Objective Establishing the model of oxidative damage in vascular endothelial cell(VEC)which are induced by oxidized low density lipoprotein(ox-LDL) to investigate the protective effects and possible mechanism of serum and plasma containing modified Buyang Huanwu Decoction(Buyang Huanwu Decoction,BHD), then providing theoretical basis for prevention and treatment of AS by Supplementing Qi and Activating Blood Circulation Method. Meanwhile,to provide an evidence for the application of plasma pharmacology in the field of Traditional Chinese Medicine,the different effects of the serum and plasma containing modified BHD on the VEC are compared.Method Using the theory of both serum and plasma pharmacology,the modified BHD-containing serum and plasma are prepared. Human umbilical vascular endothelial cells(HUVECs)are treated with serum and plasma containing modified BHD,establishing the model of oxidative damage in HUVECs which is induced by ox-LDL(150ug·ml-1)for an additional 24 hours. By assaying the proliferation ability of HUVECs using MTT assay and the HUVECs apoptosis are with Hoechst 33258 staining and flow cytometry, meanwhile,determining the level of nitric oxide(NO)、 malondialdehyde(MDA) and the activity of superoxide dismutase(SOD)、glutathione peroxidase(GPx), the protective effect of serum and plasma with modified BHD on injured VEC are assessed,so are they in comparing the differences between serum and plasma containing modified BHD.Result The percentage of cells death and apoptosis are significantly higher in the model group than that in the control group(P<0.05).As compared with the model group,results of the modified BHD-containing serum and plasma group showed that cell viability is significantly increased (P<0.05),the level of NO release, as well as the activities of SOD and GPx are all signifificantly higher,the level of MDA release are significantly lower (all P<0.05). Besides, the percentage of cells death and apoptosis are lower in modified BHD-containing serum group than that in plasma group(P<0.05). However,the level of NO release in modified BHD-containing plasma group are higher than that in serum group(P<0.05),the level of MDA and the activities of SOD、GPx between modified BHD-containing serum and plasma are no significant difference(P>0.05). Conclusion Supplementing Qi and activating blood circulation method can be one of the main therapeutic principle and methods of AS.The serum and plasma containing modified BHD could prevent VEC from oxidative injury induced by ox-LDL and improving the function of endothelial cells、anti-oxidative damage、inhibiting cell apoptosis may be its mechanism of preventing. Under certain conditions,plasma pharmacology is better than the method of serum pharmacology.
