Objective:To explore the difference of expression of BRCA1,FANCF,CD44+CD24-which is phenotypic markers of breast cancer stem cells in triple negative and luminal B (Her-2 positive) breast cancer.To explore basis for evaluating prognosis of triple negative breast cancer.Methods:1. Select 29 paraffin-embedded tumor specimens of triple negative breast cancer and 28 of luminal B(Her-2 positive)breast cancer from patients in breast surgery from the year of 2013 to 2015 who are not treated by chemotherapy before operation or biopsies.2.Slice the paraffin specimens and detect the expression of CD44 protein, CD24 protein,BRCA1 protein and FANCF protein by the method of immunohistochemistry.3.Through the software SPSS 19.0, the differences of BRCA1,FANCF,CD44+CD24-phenotype between two groups were analyzed by the method of x2 test,the correlations between CD44+CD24- phenotype and the expression of BRCA1 and FANCF protein were analyzed by the method of Spearman correlation.Results:1. In the group of triple negative breast cancer,the positive rate of CD44+CD24-phenotype was 89.7%(26/29).The positive rate of BRCA1 protein was 6.9% (2/29).The positive rate of FANCF was 75.9%(22/29).While in the group of luminal B (Her-2 positive) (simply’triple positive’for short below) breast cancer, the positive rate of CD44+CD24-phenotype was 67.9%(19/28).The positive rate of BRCA1 protein was 28.6%(8/28).The positive rate of FANCF was 75%(21/28)2. There was statistical difference of the expression rates of CD44+CD24-phenotype between two groups(P=0.044). There was statistical difference of the expression rates of BRCA1 between two groups(P=0.031).There was no statistical difference of the expression rates of FANCF protein (P=0.940)3. The loss rates of BRCA1 were consistent with the positive rates of CD44+CD24-phenotype in both two groups.The results of pearson correlation reveals that expression of CD44+CD24- phenotype was relevant to BRCA1 protein (P=0.013),while not relevant to FANCF protein (P=0.147)Conclusions:1.There was statistical difference of the expression rates of BRCA1 between two groups(P=0.031).2.There was statistical difference of the expression rates of CD44+/CD24-phenotype between two groups (P=0.044)3.While there was no statistical difference of the expression rates of FANCF protein (P=0.940).4.There was statistical correlation between CD44+/CD24-phenotype and expression of BRCA1 (P=0.013).However,the expression of FANCF was not related with CD44+/CD24-phenotype (P=0.147)5.The results revealed that mutation rate of triple negative breast cancer was higher than triple positive breast cancer,in addition,the mutation of BRCA1 may contribute to the increase of breast cancer stem cell.The role of FANCF in the development of triple negative breast was undiscovered.
