| Objective:To evaluate the effect of Shenlingbaizhu powder on intestinal mucosal barrier in immature rats with spleen deficiency type diarrhea, which is based on the clinical application of shenlingbaizhu powder and the close link between the spleen deficiency diarrhea with the damage of intestinal mucosal barrier. To clarify the possible mechanism of Shenlingbaizhu powder in treating spleen deficiency of chronic diarrhea. In order to shorten the time of treatment in children and reduce the medical expenses, even the mortality rate of children with chronic diarrhea in our country and the world. In hope that Chinese medicine make due contributions to the cause of global child health care.Methods:72 Wistar immature rats are randomly divided into six groups:blank control group, model control group, low, medium and high dose group of Chinese medicine, western medicine control group. Making spleen deficiency diarrhea rat models by intragastric administration of rhubarb decoction, and then give the drug. Testing the pathological changes of colonic tissue in rats by light microscopy and ultrastructure of colon by electron microscope. Detecting the ratio of fructose to mannitol (L/M) in urine of rats. Meanwhile, determining the changes of plasma D-levels of lactic acid, two amine oxidase and expression of proliferating cell nuclear antigen (PCNA). Finally, to process the statistics and analysis.Results:The experiment successfully replicates rat model of spleen deficiency syndrome. The diarrhea rate and diarrhea index of the experimental group are significantly different from the control group (P<0.01). So the model is successful. The light microscope observation shows: juvenile rats intestinal does not exist obvious organic disease. Intestinal mucosa changing is not significant before and after the treatment. The electron microscope shows:Before treatment, the animal model of colonic mucosal epithelial microvilli on the surface of significantly reduced, shorter, loosely arranged, even missing.Part of intestinal mucosal cell nuclei pyknosis, swelling of mitochondria, cell matrix density reduce, crest reduction.After two weeks of treatment, microvillus growth of Shenling Baizhu powder, each group rats were improved significantly. Compared with the control group, different doses of shenlingbaizhu powder can reduce the ratio of urine in rats of L/M, plasma D-levels of lactic acid, DAO and the middle and high dose group could significantly improve the proliferation cell ratio and increase the number of positive cells in the PCNA expression in the intestinal crypt area.Conclusion:Spleen deficiency diarrhea rats present obvious intestinal mucosa structural damage and functional decline. Shenlingbaizhu powder can significantly reduce intestinal mucosal injury and decrease the intestinal permeability in model rats. Shenlingbaizhu powder has a protective effect on intestinal mucosal barrier in rats with spleen deficiency diarrhea. |
