Ever since a long time ago, glioma is a troublesome for neurosurgery doctors, iteration hospitalizations and chemotherapies is a very very long and repeatedly course.The pressure of psyche and economy is heavy and burden is serious. Abundant of therapeutic measures paied,such as immunofluorescence microscope and hydrogen MRS, Prognosis is very poor and affected by much more factors. The survival rate only 2-3 year for the stage Ⅲ while 1year for stage Ⅳ. Paclitaxel which is extracted from zishan, it is widely used in breast carcinoma、ovarian cancer and intracranial tumour and so on, the resistance and therapeutic effect made it became second drug. Temozolomide is a Imidazole tetrazine antitumor drug,and it is used very well at present clinical practice,therapeutic response decreased and drug resistance become much more obviously. The resistance mechanism becomes more clear accompany the penetrate deeply researched. with the purpose of prolongation the living time and exaltation the living quality, amelioration drug resistance to become chief matter. Is PTX coadunats TMZ could educe synergetic effect?synergistic reaction is obviously in our research. Temozolomide combine with Paclitaxel coule become a optimization of labor organization in the future. Objective:The purpose of this experiment is demonstrates peclitaxel could ameliorate the sensitivity of temozolomide in vitro, and provided the rationale for koinonia.. Methods: 1 Preparation experiment:preparation glioma cell of U87,related reagent and materials,Reading correlative literature and experiments mechanism,to have an intimate knowledge of experiments technology. 2 Cell cultured:learning the knowledge of cell cultured,comprehend the vegetative cycle ofU87,morphology characteristics and habitus of Biology. 3 Draw cell multiplication arcuation used CCK-8 about paclitaxel combine temozolomide,determine generation inhibition ratio. 4 Use SPSS11.0 to give statistics handling. Results:PTX、MTZ and the combination both can inhibites the generation activity,concerned with time and dosage. the two down regulates the MGMT and MPG ameliorates the sensitivity of TMZ. Conclusion:The synergetic effect is much obviously in PTX and TMZ group, the effort is correlates to time and dose.
