Objectives and Backgroud:To investigate the inhibition and mechanism of Temozolomide(TMZ) and PTX are very effective drug for malignant gliomas with significant survival increase.Preclinic and clinic investingations have demonstrated that the combination therapy of TMZ with PTX on glioma in vitro are empty. Our findings not only suggest the original TMT Paclitaxel in combination with great potential as a cancer therapy application, we investigate the effects of lidamyeinonra t glioma C6 cell line Materials and Methods:The vitro sensitivity of drug were studied with cell culture, Proliferation rate of C6 glioma cells in each group was detected by MTT assay. FCM detected apoptosis and mitochondrial membrane potential of C6 glioma cells in each group. RT-PCR detected Caspase-3 mRNA expression in each group.Results:The results of vitro experiment:Compared with the NC group, in PTXGroup, TMZ Group and PTX+TMZ Group, the proliferation rate of cells were decreased(P<0.05); Compared with the NC group, the apoptosis rate, upregulates the ratio of Caspase-3 expression to promote the apoptosis of the C6 glioma cell.(P<0.05).Conclusions:Our experiments revealed that combination chemotherapy of TMZ and PTX can obviously inhibit the proliferation and induce the apoptosis in glioma cell and futheremore the mechanisms were initially analyzed. It supplied the experimental and theoretical basis for the combination chemotherapy in treating glioma in clinic in future.
