| Objective:This study aimed on the probiotic features of Lactobacillus salivarius w22 a and it’s effects against peri-implantitis pathogens in vitro.In order to provide experimental basis of Lactobacillus salivarius w22 a as probiotics in the prevention and treatment of peri implantitis via the experiments.Methods:1.Preliminary analysis of metabolites of Lactobacillus w22a:Measurement of pH values of metabolites of,using Streptococcus as a control;The content of hydrogen peroxide was detected by semi quantitative test paper;The content of extracellular polysaccharide was measured by phenol sulfuric acid method.2.Measurement of the rate of self polymerization of Lactobacillus w22 a and coaggregation with pathogenic bacteria by using turbidity method.3.To observe the inhibitory effect of Lactobacillus w22a’s metabolites on Pgã€Piã€Fn by using the agar diffusion method.4.To observe the tolerance of Lactobacillus salivarius w22 a to lysozyme and antibiotics by using agar diffusion method and co culture method.Results: 1.Preliminary analysis of the fermentation of Lactobacillus salivarius w22 a.Lactobacillus salivarius w22 a could produce organic acid,and acid producing ability was weaker than that of Streptococcus mutants; A certain amount of hydrogen peroxide and extracellular polysaccharide also could be examed.It was suitable for using Lactobacillus salivarius w22 a as probiotics besause of its metabolic characteristics.2.Aggregation experiment showed that Lactobacillus salivarius w22 a got auto-aggregation and co-aggregation abilities with Porphyromonas gingivalis(Pg) 〠Pevotellaintermedia(Pi) ã€Fusobacteriumncleatum(Fn);besides,Lactobacillus salivarius w22 a was hydrophobic.The adhesion characteristics might help its colonization in the dental implants environment.3.By applying fermentation of Lactobacillus salivarius w22 a to antibacterial experiments in vitro,it was showed that the fermentation could inhibition the growth of Pgã€Piã€Fn in different degrees.It could be concluded that the inhibition was a complex function of many substances in its fermentation.4.The tolerance of antimicrobial substances was evaluated.Lactobacillus salivarius w22 a exhibited tolerance to some kinds of antibiotics.The strain also showed tolerance to 0.8mg/mL Lysozyme solution.It was good for its survival in the oral cavity at some degree.Conclusion: Lactobacillus salivarius w22 a is a strain with potential oral probiotic features and expected to be used as probiotics in the field of prevention and treatment of PI. |
