| Objective:This study through established the model of rats with severe abdominal infection,comparing the activity of diamine oxidase(DAO) and the concentration of D-lactic acid in plasma,and the pathological change of terminal ileum were observed and graded under optical microscope,to explore the possible protective effect of taurine on intestinactionl mucosal barrier function in rats with severe abdominal infection.Methods:Because a higher mortality was expected with cecal ligation and puncture(CLP),Sixty-six SD males rats were randomly divided into three groups:control group(Group C n=18),abdominal infection group(Group I n=24)and taurine treatment group(Group T n=24).The rats of group C were only slightly changed the cecal several times before definitive abdominal closure.The rats in group I and group T were established by CLP.The rats in group T received intraperitoneal injection of taurine(200mg/kg)immediately after the operation,however,the rats in group C and group I received intraperitoneal injection of equivoluminal normal saline.Six rats in group C were randomly picked up at 6,12,24 h after operation,eight rats in group I and group T were randomly picked up at 6,12,24 h after operation to have the activity of diamine oxidase(DAO) and the concentration of D-lactic acid in plasma measured.The pathological change of terminal ileum were observed and graded.Results:(1)Serum DAO activity: Main effect of both grouping factors and time factors had statistically significant(P<0.05),and there was no interaction between grouping factors and time factors(P > 0.05).According to the consequence of pairwise comparison,the comparison between different groups and different time had statistically significant(P<0.05).The activity of DAO was significantly higher of the group I than that of group T,which was higher than that of group C at the same point of time(P<0.05).(2)Serum D-lactic acid concentration:Main effect of both grouping factors and time factors had statistically significant(P < 0.05),and there was no interaction between grouping factors and time factors(P> 0.05).According to the consequence of pairwise comparison,the comparison between different groups and different time had statistically significant(P<0.05). The concentration of D-lactic acid was significantly higher of the group I than that of group T,which was higher than that of group C at the same point of time(P<0.05).(3) Pathological injury classification of ileum mucosa:Main effect of both grouping factors and time factors had statistically significant(P < 0.05),and there was no interaction between grouping factors and time factors(P > 0.05).According to the consequence of pairwise comparison,the comparison between different groups and different time had statistically significant(P < 0.05).The grade of pathological injury classification of ileum mucosa was significantly higher of the group I than that of group T,which was higher than that of group C at the same point of time(P<0.05).Conclusion:Taurine has a certain protective effect on intestinal mucosal barrier function in rats with severe abdominal infection. |
