Objective:Developed the non-add PCDD/Fs and DL-PCBs reference material of beef, strengthen quality control of laboratory of the dioxins’ reference materials in food and filled blank of the dioxins reference materials in food. Used the prepared reference material to analysis PCDD/Fs and DL-PCBs in beef and milk which got from market. In addition, we used the pollution data to assess people’s dietary exposure preliminarily in order to provide basis data for further study of diet. Methods:Selected the higher detection rate of dioxins in beef samples, through homogenization,freeze-dried, splintered, sifting, blending, and packaged the powder in brown bottles.PCDD/Fs and dl-PCBs in samples were quantified by HRGC-HRMS, using isotope dilution methodology.Analysis of variance(ANOVA)and F test were used for homogeneity, simple linear regression was used with stability, there are 9 laboratories collaborated to setting the value of the reference material uncertainty. Results:Produced reference material according to the JJG 1006-94, 10 kinds of PCDD/Fs and 11 DL-PCBs’ F values were less than F0.05(14,30),2378-TCDD,2378-TCDF,12378-Pe CDF,123789-Hx CDF,123478-Hp CDF、OCDF were not detected, the prepared reference material was well uniformed. After a year for long-term stability test at room temperature,PCDD/Fs and DL-PCBs of reference material, it was proved not have long-term variation trends.21 kinds of dioxins ’ value was12378-Pe CDD:0.12±0.1pg/g; 123678-Hx CDD:0.20±0.07pg/g; 123789-Hx CDD:0.06±0.05pg/g;1234678-Hp CDD:0.6±0.3pg/g; OCDD:1.4±0.6pg/g; 23478-Pe CDF:0.4±0.2pg/g;123478-Hx CDF:0.19±0.06 pg/g; 123678-Hx CDF:0.18±0.06 pg/g; 234678-Hx CDF:0.12±0.03 pg/g;1234678-Hp CDF:0.1±0.2pg/g; PCB77:0.6±0.4pg/g; PCB105:12±6pg/g; PCB114:4±1pg/g;PCB118:37±16 pg/g; PCB123:1±1 pg/g; PCB126:1.8±0.6pg/g; PCB156:11±4pg/g; PCB157:4±2 pg/g;PCB167:5±2 pg/g; PCB169:1.3±0.6 pg/g; PCB189:5±2 pg/g.Used the prepared reference material to analysis dioxins in beef and milk bought from market. The lever of dioxins in beef samples ranged from 0.06 pg TEQWHO-05/g fat to 56.59)pg TEQWHO-05/g fat, with a median of 1.93 pg TEQWHO-05/g fat. In the current polluted condition to estimate the maximum intake lever of dioxins through beef was 6.54 pg TEQ/kg bw/month, accounted for 9.2% of PTMI.Dioxins in cow milk in the range of(0.45 ~ 15.21) pg TEQWHO-05/g fat,the average level is 3.81 pg TEQWHO-05/g fat. Residents’ intake levels of dioxins were(1.87 ~ 49.32) pg TEQ/kg bw/month through milk. Accounting for 3% ~ 70% of PTMI.Conclusions:The study was prepared 150 bottles reference material of dioxins in beef Dioxins total of 150 bottles of standard material, the homogeneity and the stability were meeting the requirements of standard, the collaboration of 9 laboratories setting results. To provide standard and quality control reference for analysis of dioxins in food. There were different levels of dioxins pollution in beef and milk, there were some differences with existing food samples’ and environmental samples’ PCDD/Fs contamination contour. Overall, the levels of dioxins were below the EU about the maximum limit of beef and milk. Residents of a low level of beef consumption, so the dietary risk level was low through by the beef, in spite of the high pollution in some beef samples. Simultaneously milk consumption level were different greatly, dietary exposure situation is different, there were some health risks for the people had high consumption of milk.
