Objective To elucidate sustained-release preparations in vitro release and improved the relationship between the properties of peach gum, polysaccharide components;Methods(1) Through the determination of solubility method to determine the original peach gum and to improve the solubility of peach gum;Use weighing method,determination of the original peach gum and improvement of peach gum swelling and wet weight, dry weight, calculate the equilibrium swelling ratio;Using rotary viscosity measurement method for determining the dynamic viscosity of the original peach gum and improvement of peach gum;Properties such as solubility, viscosity, the equilibrium swelling ratio as an index, study the properties of the peach gum improved before and after different regions.(2) Using Moish reaction, Fiji Lin, phenol- sulfuric acid reaction, 3, 5-2 nitro salicylic acid, KI- I2, biuret test, ninhydrin test, such as general chemical identification methods to identify different origin peach gum improved chemical composition before and after.Using phenol- sulfuric acid method for determining the polysaccharide content of peach gum improved before and after;Through acetylation gas chromatography at the same time, before the PMP column derivative HPLC analysis of different regions peach gum polysaccharide composition before and after improvement.(3) Different origin peach gum out of flat peach gum and improvement zyban winning component ephedrine and pseudoephedrine, the influence of scopolamine release property in vitro.Results(1) the solubility of peach gum different regions for 4 ~ 9%, the equilibrium swelling ratio of 17 ~ 35.16 g/g 1, viscosity is 19 ~ 48.4 m Pa s., percentage of polysaccharide content is 69 ~ 73%, arabinose, galactose, and xylose in peach gum polysaccharide than 25 ~ 30% respectively, 20 ~ 25%, 4 ~ 6%, asthma drug controlled-release film in 1 ~ 2 h released in full.(2) Peach gum improved solubility is 6 ~16%, the equilibrium swelling ratio of 40 ~ 67 g/g 1, viscosity is 105 ~ 287 m Pa, s,percentage of polysaccharide content is 75 ~ 85%, arabinose, galactose, and the ratio of xylose in peach gum polysaccharide, respectively is 33 ~ 40%, 25 ~ 30%, 10% ~ 30%, out of slow-release drug film in 6 ~ 12 h released in full.(3) Improvement of peach gum polysaccharide content and the higher the percentage, the greater the viscosity, solubility and swelling properties, the better, the preparation of drug release and the longer zyban.(4)Improved the properties of peach gum, polysaccharide content, different regions out of flat sustained-release film in vitro release, the optimal, guizhou guiyang reformed peach gum polysaccharide, the highest percentage content out of flat zyban release rate index constituents in the slowest;The worst origin for jilin gongzhuling.Conclusion modified high percentage of peach gum polysaccharide content, viscosity, in vitro drug release rate is slow, to study for Chinese traditional medicine complex composition equilibrium release of accessories to provide the reference. |
