| Endometriosis is common diseases in reproductive age women. the incidence is about 10% ~ 15%, with increasing trend. Secondary dysmenorrhea is the most typical clinical symptoms, 50% of patients is combined with infertility[1]. Its biological behavior,similar with malignant tumor, involving peritoneum, ovaries, fallopian tubes, bladder and so on. The malignant transformation rate is 0.33% ~ 1.5% [2], [3].The level of serum carbohydrate cancer antigen 125,199(CA125 and CA199) have diagnosis value. They have high expression in uterine epithelial tissue and are used to evaluate[4]. From the perspective of tumor, studies confirm the relationship between tumor and abnormal cell cycle. Meanwhile studiers have explored association, and speculate that the development is leaded by abnormal cell cycle protein expression and proliferation. Cell cycle protein D(CyclinD) is positive factor in the cell cycle. By combining with the cell cycle protein dependent protein kinase CDK4 and CDK6 through G1 checkpoint and S phase [5]. Through competition with CyclinD1(a suptype of CyclinD), P16 combine with CDK4 and CyclinD1 CDK4-complex, to suppress the activity of CDK4 and prevent G1 to S phase and DNA synthesis. It play an role as a negative factor[6], related to abnormal growth, proliferation, infiltration and metastasis of ectopic endometrium and malignance.Professor Cao Baoli studied the effects of prescription compound Rhizoma Curcuma powder on endomentriosis. We choose serum CA125, CA199, CyclinD1 and P16. We examined the level of CA125 and CA199 and expression of both CyclinD1 and P16,analyse the basic theory of its treatment on endomemtriosis. Objective:1.To observe the level of serum CA125, CA199, explore curative effect of compound Rhizoma Curcuma powder.2.To observe both CyclinD1 and P16 expression in eutopic and ectopicin endomtrium tissu in endometriosis patients, to explore its significant effection in both occurrence and development of disease and its mechanism and possible assistant curative effects during the treatment of this disease. Method:1.From January 2013 to January 2014, 69 endometriosis patients were advocated. Administration group contains 35 patients, diagnosed with endomentriosis and administrated compound Rhizoma Curcuma powder for three months before surgery; observation group includs 34 cases, without treatment; meanwhile control group includs 30 patients. Detect serum CA125 and CA199 levels before surgery and expression of cyclinD1 and P16 in ectopic endometrium and eutopic endometrium. Result:1.Both of the level of CA125 and CA199 in patients in Administration group and observation group are significant than in control group(P<0.05). In administration group, they were significantly higher lower than in observation group(P<0.05).2.CyclinD1 expression in ectopic endometrium in administration group was lower than in observation group without significance(P>0.05). CyclinD1 expression in eutopic endometriumin administration group was significantly lower than in observation group(P<0.05). CyclinD1 expression in both administration and observation group are significantly higher(P<0.05). P16 expression in ectopic endometrium in patients in administration group was higher than in observation group with significance(P<0.05). P16 expression in eutopic endometrium in administration group was significantly higher than in observation group(P<0.05). Coclusion:1.In ovarian endometriosis patients,serum levels of CA125 and CA199 is significantly higher, The compound Rhizoma Curcuma power can induce them.2.In ovarian endometriosis patients, CyclinD1 expression is high and P16 is low. It’s suggested that CyclinD1 and P16 contribute to endometriosis. Compound Rhizoma Curcumae power can significantly lower the expression of CyclinD1 in ectopicin endometrium in endometriosis patients and increase the expression of protein P16 in eutopic and ectopic endometrium in endometriosis patients. It’s suggested its role in the inhibition of cell cycle progression. It also provided a theoretical support for therapy and prevention of recurrence for endometriosis. |
