| Objective: To investigate the effects of BMP-9 gene overexpression on glucose and lipid metabolism in mice.Method: Eighty male C57BL/6J mice aged 8 week were randomly divided into two groups fed with normal diet(ND group,n=40)or high fat diet(HFD group,n=40)for 16 weeks. At the end of the 15 th week,all mice were injected with Ad-GFP or Ad-BMP-9 by tail vein and divided into NC+ Ad-GFP group(NF group,n=20),NC+Ad-BMP-9 group(NB group,n=20),HFD+ Ad-GFP group(HF group,n=20)and HFD+Ad-BMP-9group( HB group, n=20).The glucose-lipid metabolism and insulin sensitivity were evaluated by intraperitoneal glucose tolerance test,insulin tolerance test and hyperinsulinemic-euglycemic clamp technique.Results: After normal or high-fat diets,body weights in HFD group were significantly increased compared with ND group(P(27) 0.01). The BMP-9 m RNA and protein expression of mice in HFD group were significantly decreased compared with ND group(P(27)0.01). Compared with HF group, HB group has a significant reduction in body weight,foodintake, liver weight/body weight, FBG, FIns, TG, TC andFFA(P(27)0.01),while NB group decreased slightly than NF group. There were no significant differences between NF group and HB group of energy expenditure, but decreased in NB group compared with NF group(P(27)0.01). However,there were no significant differences between NF group and NB group,HF group and HB group in epididymal fat weight/body weight, heart rate and systolic pressure,respectively. During the steady-state of clamp, compared with NF group, NB group had a significant increasing in GIR and GRd,but HGP and suppression decreased slightly,while GIR and GRd of HB group increased significantly than of HF group,and the hepatic glucose production was suppressed significantly(P(27)0.05).Conclusion: Insulin resistance can be induced by high fat diet in mice. BMP-9 has an effect on high-fat diet induced insulin resistance which can improve the glucose and lipid metabolic disorders. |
