Effect And Mechanism Of Yuxiao San Combined With Cisplatin Injection By External Application On LEWIS Lung Cancer In Mice

Background: The current efficacy treatments of lung cancer, including chemotherapy, surgery and molecular targeted therapy, are still unsatisfactory. Moreover, the survival rate of patients have no significantly breakthrough. Clinical studies have shown combination of traditional Chinese and Western medicine treatment can significantly improve the survival rate of lung cancer patients and prolong survival. Yuxiao San powder is our effective prescription for the external treatment of cancer. It has great advantages in the treatment of advanced lung cancer after a long-term clinical practice.Objective: Study the effects and mechanism of Yuxiao San in inhibiting the growth of lung cancer, by measuring volume, weight and metastases of Lewis lung cancer in mice, and detecting the expression gen of K-ras and nm23 by using the immunohistochemical, q RT-PCR and Western blot.Methods: Establish of Lewis lung cancer models transplanted in C57BL/6J mice. On the day 7th of inoculation, randomly divided the mice into model group, cisplatin group(0.002 g·kg-1), Yuxiao San group(10 g·kg-1), and the combination group(10 g·kg-1/0.002 g·kg-1). Each group was consisting of 8 mice. The mice were treated by Yuxiao San powder topically 1 time a day, for 21 days. On day 29 th the mice were executed to death. Then each tumor’s weight and volume was measured, using immunohistochemical, Western blot and q RT-PCR methods to detect expressions of K-ras and nm23.Results: Compared with model group, the tumor’s volume, weight and lung metastases of combination group were decreased, and expressions of K-ras through immunohistochemical, Western blot, and q RT-PCR methods were decreased(P<0.01). And compared with model group, expressions of nm23 in tumors through immunohistochemical, Western blot, and q RT-PCR methods were increased(P<0.01). Cisplatin group, Yuxiao San group and the combination group have significant different in value compared with the model group. The difference was statistically significant(P<0.01). And the using of combination of cisplatin and Yuxiao San is better than single drug(P<0.01).Conclusion: Yuxiao San powder can reduce the tumor’s volume, weight and the mechanism of Yuxiao San in inhibiting the growth of lung cancer was relative to decrease the expressions of K-ras and increase the expressions of nm23.
