Study On Correlation Of Material Foundation On Anti-Damp-Heat-Syndrome Gout Of Ermiao Wan Categorized Formula

To study the correlation of the material foundation of on Anti-Damp-Heat-Syndrome Gout of Ermiao Wan categorized formulas (Ermiao Wan, Sanmiao Wan, Simiao Wan and modified Simiao Wan) and to explore its characteristics changes, the thesis take Ermiao Wan as the basic formula, determine the fingerprint of the effective parts of Ermiao Wan categorized formulas in anti-gouty syndrome of damp and heat, compare the composition features and compatibility changes of total peaks, and analyze the correlation and compatible mechanism of the effective parts with biological effect index by statistical methods such as correlation, regression, cluster analysis and so on.The main research content as follow:First of all, we get 6 valid parts by extraction、separation and purification from Jiaweisimiao Decoction. They are alkaloids, flavone, oil, organic acids, saponins and Liq.. We combine them according to orthogonal design. Then, we get a lot of samples. Establish the fingerprints of modified Simiao Wan, and then determine HPLC, GC fingerprints of categorized formulas and other samples.Then, through comparing retention time of fingerprint peaks and the ultraviolet spectra of peaks acquired, we can confirm the ingredients of the common peaks、the ascription and the source of the chromatographic peaks. Analyze the qualitaty of common peaks by using reference substances and HPLC/MS, GC/MS to establish the material foundation of Ermiao Wan categorized formulas.In the end, adopt multiple regression analysis(enter and stepwise), correlation and cluster to look for the relation at fingerprint peaks and drug effect. Explore Ermiao Wan categorized formulas characteristics changes by comparing HPLC, GC fingerprints. In orther to explore the correlation of material foundation and biological effect, we analyze biological effect and effective parts through hierarchical cluster analysis.By statistical analysis, we abtain the effect parts and effect peaks, and establish the the correlation of material foundation. Alkaloids, saponins and Liq. have obvious effect on anti-inflamation, analgesia, decreasing serum uric, oil have obvious effect on anti-inflamation and HUVEC growth repress rate, flavone and organic acids strengthen biological effect. Ermiao Wan have effect on anti-inflamation, analgesia, decreasing serum uric and HUVEC growth repress rate, Sanmiao Wan have better effect than Ermiao, Simiao Wan have better effect than Sanmiao, modified Simiao Wan have the best effect. The results show that categorized formulas are reasonable. Conclusion: Ermiao Wan categorized formulas have same substance- foudation and there are some correlations in the compatibility changes.
