Metabonomic is systematic study of small-molecule metabolite profiles to join physiological characteristics with external stimulus,in order to acquire related metabolic pathway and biological significance.Plant metabonomic is one aspect of metabonomic that applies in plant,which mainly focuses on plant different part,planting area,growth time and external stimulation.This paper studied the germination of rapeseed and Allium macrostemon Bunge(AMB)processing by using NMR,GC-MS and metabonomic.The results provide vital information on breeding of rapeseed and AMB processing.Plant life cycle includes several period,such as germination,flower fruit,and the spread of seed.Germination is considered as the start of plant life,which decides the extension of plant.By measuring the rapeseed weight,water uptake period could be separated into 3 phases,phase A,initial quick water uptake(0 h to 12 h);phase B,a plateau phase(12 h to 36 h);phase C,further add in water uptake(36 h to 84 h).We detected 35 metabolites,including organic acid,amino acid,sugars,alkoloids,and so on.PCA and OPLS-DA showed the clear difference among the 8 groups sample,indicated that metabolism is active during the period of germination.Sugar and myo-inositol have a bearing on energy,while sugar might be the primary energy supply for rapeseed germination.The change of proline showed that it not only functioned as carbon and nitrogen,but also helped to form cell wall in phase C.The stable betaine showed its regulation for osmotic equilibrium during the rapeseed germination.Traditional Chinese herbal medicines usually need dry before storage;plant loses most of water from fresh to dry,which causes the metabolite variation.It's reported that sulfur compounds might be the active components of AMB,this paper investigated sulfur difference of AMB caused by freeze-drying,shade-drying.In this study,we detected 10 sulfides from AMB using GC-MS.They changed little on their metabolite variety,but their content in each group altered significantly.The content of sulfides from freeze-drying sample decreased integrally compared to the fresh-sample,while for the shade-drying sample,only dimethyl disulfide,dimethyl trisulfide decreased.Multivariate statistical analysis suggests that freeze-drying has little effect on sulfide of AMB compared to shade-drying,which might be caused by temperature and light during the drying processing.Therefore freeze-drying might be the more efficient way to maintain the active components,sulfides in AMB.
