Cardamine Enshiensis is a kind of newfound selenium hyperaccumulators. Exaction and Speration, preliminary purification, and antioxidant activity of selenium-containing protein from Cardamine Enshiensis were rarely reported at present. Exaction and Speration, pr Cardamine Enshiensiseliminary purification, and antioxidant activity of selenium-containing protein from Cardamine Enshiensis and the nutrient levels of Cardamine Enshiensis were studied in this paper.The conditions for the extraction and purification of selenium-containing protein from Cardamine Enshiensis and the antioxidant activity in vitro were explored, too. The major work were summarized as follows:(1)This was the first study on the systemical nutrient levels of Cardamine Enshiensis. The results showed that the content of Se was rich. Cardamine Enshiensis is rich in essential nutrients,such as water, ash, crude fat, crude ptotein, dietary fibre. There are lots of value of development and application as a newly plant resources. The work supplied the basic data and necessary theoretical guidance for further experiments.(2) Exaction and Speration, preliminary purification, and antioxidant activity of different soluble proteins from Cardamine Enshiensis were researched firstly in this paper. The main protein component is water soluble protein, accounting for 37.57% of total protein, followed by alkali soluble proteins, accounting for 35.75% of total protein. Alkaline dissolved protein had the strongest scavenging power to reducing activity and total antioxidation capacity(T-AOC). Water dissolved protein was the best to eliminating super oxygen anion free-radicals. hydroxyl. The work meaned that alkaline dissolved protein was a ideal materials for the Exaction of selenium-containing protein.(3) The extraction technology of alkaline dissolved protein from Cardamine Enshiensis and process optimization by orthogonal experiment and response surface experiment was studied firstly in this paper. The effect of factors, such as p H, extraction time, extraction temperature, and the ratio of extration solvent to material on extraction rate of alkaline dissolved prorein from Cardamine Enshiensis were investigated, respectively. The optimum extraction conditions were obtained as follows: p H 10, extraction time 3 h, extraction temperature 40 ?, solid- liquid ratio 1:40 g/m L, extraction times 2. And then the best extraction conditions are determined through the orthogonal experiment. Under such conditions the protein extraction ratio could be 48.24%. Experimental des ign and experiments of this technology were carried out. Responsesurface model with extraction rate of protein as the optimization goal was built. The optimum extraction conditions were obtained as follows: p H 11, extraction time 3.5 h, extraction temperature 40 ?, solid- liquid ratio 1:40 g/m L, Under such conditions the protein extraction ratio could be 49.68%. Process optimization of extraction of alkaline dissolved prorein from Cardamine Enshiensis offered a theoretic basis for extractio n of alkaline dissolved prorein and laid the foundation for further development and utilization of Cardamine Enshiensis.(4)This paper used Sephadex G-100 and DEAE-Sephcrose-FF to exact and purify selenium-containing protein from Cardamine Enshiensis for the first time. High concentration proteins was obtained and its antioxidant activity was determined.The results showed isoelectric point of High concentration proteins and molecular weight was 4.8 and 17~25 k Da respectively. High concentration proteins was rich in cysteine and leucine. Alkali soluble protein extract from Cardamine Enshiensis was purified to homogeneity by Sephadex G-100 gel filtration chromatography, getting 4 peaks in purified myelin.There are the largest Selenium and protein contents in peak ?. The concentration of Se was 2.52 ?g/m L, The concentration of protein was 7.85 mg/m L. Peak?was purified to homogeneity by DEAE-Sephcrose-FF, getting 4 peaks in purified myelin. There are the largest Selenium and protein contents in peak c. The concentration of Se was 1.29 ?g/m L, The concentration of protein was 3.49 mg/m L. The anti-oxidation activity of 4 peaks in purified myelin from DEAE-Sephcrose-FF were studied. The results showed peak c has the strongest hydroxyl radical scavenging activities, Superoxide radical scavenging activities and DPPH free radicals scavenging activities comparing to peak a, peak b and peak d. The more Se the fraction has, the stonger anti-oxidation activity is.The work provided essential condition for analysis o f secondary structure of selenium-containing protein from Cardamine Enshiensis...
