Objective:The dried root of Morinda officinalis How with the function of tonify kidney-Yang effect,strong bones and muscles,reduce rheumatism,is widely used in impotence,seminal emission,cold infertility,menstruation disorder,Shaofu cold sore,rheumatic pain,muscles atrophy disease and so on in southern China.It has been as the main medicinal parts was collected in the Chinese Pharmacopoeia.Morinda cochinchinensis DC.,Morinda parvi folia Bartl.ex DC.and Morinda umbel lata L.subsp.obovata Y.Z.Ruan also have treatment of rheumatism and other drug treatment.And The fruit of Morinda citrifolia L.was also found to have significant effect on varieties of diseases,such as antiviral,anti-cancer,anti rheumatic.The above plants all are the plant species of genus of Morinda L.,thus,plants belong to the genus of Morinda L.have greater medicinal potential.This study aims at finding a suitable identificated way to Morinda officinalis How and its related six plants,and to provide the necessary basis for the breeding of Morinda officinalis How and its six kinds of close relatives,so as to reduce the situation of true and false chaos of medicinal plants caused by wrong planting,wrong harvest and wrong collection,and ensure that the medicinal plants Pure varieties.The above six related plants are Morinda umbel lata L.subsp.obovata Y.Z.Ruan,Morindaparvi folia Bart1.ex DC.,Morindacochinchinensis DC.,Morinda hainanensi s Merr.et How,Morinda ci tri folia L.and Morinda badia Y.Z.Ruan.Methods:1.Based on the image analysis technique,the basic shape features data of the leaves in the plant leaf image were measured and collected,and the basic shapefeatures of the leaves were transformed into a further description features of the leavesshape.Identification of experimental species based on shapecharacteristics of leaves using systematic cluster analysis.2.Based on the image analysis technique,the basic microscopic characteristics of non-glanduLar hairs in the non-glanduLar hairs image were measured and collected.Identification of experimental species based on microscopic characteristics of non-glanduLar hairs using system clustering analysis.3.The Fourier transform infrared spectroscopy and system cluster analysis were used to identify the experiment related plant species.The infrared spectra of the experimental samples were obtained,and the experimental resuLts were classified and identified based on the characteristics of infrared spectroscopy.4.The DNA barcode sequences ITS2,psbA-trnH,matK,rbcL and trnL-trnF were amplificated from the genomic DNA of the leaves of the experiment related plant species.Genetic diversity based on sequence alignment,similarity genetic distance and maximum likelihood system development tree evaluate the identifiction ability of the 5 DNA barcode sequences for experiment related plants species.The DNA barcode sequences suitable for identification of experimental species were screened after comprehensive evaluation of the species identification ability of each sequence.5.All the genes of Morinda officinalis How were downloaded from Genbank database,and the functions of the gene sequences was enriched by gene set.The single gene related to DNA barcode sequence in Morinda officinalis How genes was annotated.The morphological characteristics of plant leaves morphology,non-glanduLar morphology,chemical composition and DNA barcode sequence were compared with the gene function information of Morinda officinalis How to explore the intrinsic relationship between plant leaf morphology,non-glanduLar morphology,chemical composition and genetic information.ResuLts:1.Based on the leaf shape characteristics,three kinds of plants which are Morinda officinalis How,Morinda umbel lata L.subsp.obovata Y.Z.Ruan and Morinda citrifolia L.were identified.2.Based on the microscopic characteristics of leaf non-glandular hair,all seven kinds of plants which are Morinda officinalis How,Morinda umbellata L.subsp.obovata Y.Z.Ruan,Morindaparvifolia Bart1.ex DC.,Morindacochinchinensis DC.,Morinda hainanensis Merr.et How,Morinda citrifolia L.and Morinda badia Y.Z.Ruan.were identified.3.Based on the infrared spectra of leaf chemical constituents,three kinds of plants which are Morinda officinalis How,Morinda hainanensi s Merr.et How and Morinda ci tri folia L.were identified.4.Based on the molecular character of DNA bar code sequence,the three kinds of plants which are Morinda offi cinali s How,Morinda hainanensis Merr.et How.and Morinda ci tri folia L.were identified.And the ITS2 sequence was selected as the optimal bar code sequence to identify the experimental species.5.The function of Morinda officinalis How genes in Genbank database is mainly related to the process of plastid,chloroplast,membrane,thylakoid membrane,transmembrane helix and transmembrane of plant leaf.Among them,psbK,psbA,psbA,atpF and atpH genes are associated with plant photosynthesis pathways.The trhH-GTG gene is associated with histidine metabolic pathway.The rpoB gene is associated with purine metabolic pathway,pyrimidine metabolic pathway and RNA polymerase.The rbcL gene is associated with B Uric acid and dicarboxylic acid metabolic pathway,photosynthetic carbon fixation pathway and carbon metabolism pathway are related.Conclusion:The non-glandular hairy microscopic characteristics of the leaves were the most effective features of Morinda offi cinalis How,Morinda umbellata L.subsp.obovata Y.Z.Ruan,Morindaparvifolia Bart1.ex DC.,Morindacochinchinensis DC.,Morinda hainanensis Merr.et How,Morinda ci trifolia L.and Morinda badia Y.Z.Ruan.,which could successfully identify all the above species,Leaf shape characteristics,leaf chemical composition infrared spectroscopy and DNA bar code sequence molecular characteristics of the species identification ability is not ideal,can only identify three kinds of experimental species,in which the chemical composition of leaf infrared spectroscopy and DNA bar code sequence molecular characteristics can identify the species Is the same,the correlation between these two features is large.
