The Heme's pheasant(Syrmaticus humiae)is a rare and endangered species of pheasants in the world that used to be widely distributed in the mountain areas.This wild population size of this species has been decreasing dramatically in rencent years.Therefore,it was listed as a globally near-threatened species by IUCN and ranked as Grade I National Protected Wildlife in China and was also listed in Appendix I of the Convention on International Trade in Endangered Specicies(CITES).The main results are as follows:This study is based on the Syrmaticus humiaes,which are wild population and captive population,genetic relationships of Syrmaticus humiaes and endangered pheasants,located in Dazhongshan National Nature Reserve,Yunnan Province,were determined at the molecular level,which provide molecular evidence for the migration of Syrmaticus diffusion pattern and habitat selection.And assess the current status of Hume's pheasant germplasm resources at molecular level,which provide scientific basis for the reasonable protection,sustainable utilization and genetic improvement of germplasm resources.(1)DNA was amplified from 8 captive and 13 field individuals,respectively,by 100 random primers,which found 7 primers amplified bands with good repeatability,clear bands,strong specificity and abundant numbers.The reaction components and reaction procedures were screened to enhance the stability of the ISSR-PCR reaction before the PCR reaction.(2)The results of this study is calculated,by the software of Popgene,the number of alleles detected Na= 1.9072,the effective number of alleles Ne= 1.5531,polymorphism rate reached 90.72%,Nei's genetic diversity index(He)is 0.3251,Shannon's information diversity index(I)is 0.4844,total gene diversity(Ht)is 0.3123,the genetic diversity within population(Hs)is 0.2787,genetic differentiation(GST)is 0.1077,gene flow(Nm)is 4.1446,the range of genetic distance is 0.25-0.44 by statistical software of 21 Syrmaticus humiaes.Syrmaticus humiae have higher genetic diversity,and studies have shown that Syrmaticus humiae captive population genetic diversity level significantly lower than the level of population genetic diversity of wild.All the above indicators are indicated that Syrmaticus humiae groups have higher genetic diversity.(3)By using Ntsys software to calculate the genetic similarity coefficient between individuals,Syrmaticus humiae 21 clustering analysis,preliminary understanding the genetic structure of the Syrmaticus humiae,which provided a new basis for the field protection and artificial breeding of the Syrmaticus humiae.
