Amaranthus caudatus L.named as lao qiang gu,xian ren gu,is a perennial herbage of A.caudatus of Amaranthaceae.It originates from tropical and subtropical zone and wide spread in the word wildly.It cultivates in all parts of China,it grows in the farmland,aside the ground,around the residence and so on.A.caudatus has a long edible history;it is not only a kind of vegetables but also a commonly Chinese herbal medicine as a folk medicin,the whole grass and seed of A.caudatus can be used for clearing away heat on detoxifying,clearing liver and making eyes bright,cooling blood,dispelling turgescence and nourishing lustihood.Although A.caudatus has commonly used as a chinese herbal medicine in some part of China nationality areas,further researches in chemical components,clinical and pharmacology activity are very little.In order to explore medicinal and edible plant,the anti-oxidative,anti-bacterial,anti-tumor and anti-diabetic activities of crude extract of effective parts of A.caudatus were determinated.The results as follow:1.Acitve screeningDifferent fraction extracted by petroleum ether,ethyl acetate,n-butyl alcohol,the active fraction which were determined by anti-oxidant,anti-bacterial,anti-tumor and anti-diabetic evaluation in vivo.The result of anti-oxidative activities showed that n-butyl alcohol crude extract has the best scavenging capacity of DPPH·(IC50=9.1189 mg/mL);ethyl acetate crude extract has the best scavenging capacity of hydroxyl free radical(IC50=1.4060 mg/mL)and the inhabition capacity of oxidative damage to deoxyribose(IC50=0.02578 mg/mL);n-butyl alcohol and ethyl acetate crude extract did not show anti-tumor activities,but petroleum ether and water crude extract showed a low anti-tumor activities evaluation after 48h;70%ethanol rude extract did not show anti-tumor activities in vitro and anti-diabetic activities on rats.ethyl acetate and petroleum ether crude extract showed a high anti-bacterial activities.2.Separating of active chemical constituentsEleven compounds were separated from from n-butyl alcohol and ethyl acetate rude extr-act of A.caudatus through extraction and purification.The eleven compounds' structure were identified by 1H-NMR,13C-NMR,MS and their physical propeties.They were(1)Hexadecanoic acid,(2)a-spinasteryl,(3)a-spinasteryl ?-D-glucopyranoside,(4)uracil,(8)trans-ferulic acid(9)uridine,(11)zarzissine,(12)thymidine,(13)quercetin-3-rutinoside,(14)luteolin 7-O-glucoside,(15)luteolin.3.In this paper,low polar fractions were extracted by steam distillation and identified by GC-MS method.4.The total flavnoids of A.caudate were determinate by colorimetric method.5.Active screening for the chemical constituents.Screened of anti-oxidative activities of A.caudatus by scavenging 1,1 diphenylhydrazlfree radical(DPPH·).Luteolin,luteolin 7-O-glucoside,quercetin 3-rutinoside exhibit show strong scavenging DPPH free radical ability:Luteolin(IC50=0.0360 mg/mL)>luteolin 7-O-glucoside(IC50=0.1546 mg/mL)>quercetin-3-rutinoside(IC50=0.4048 mg/mL).When the concentration of luteolin reached 1.00×10-1mg/mL,the inhabition capacity of human hepatoma cell lines HepG2 and SMMC-7721 reached 81.31%,76.42%respectively.The research suggested that flavonoids are the most important substance for anti-oxidation from A.caudatus.
