| Aflatoxin is a secondary metabolite produced by the fungus aspergillus flavus, which caused a lot of damage for humans and animals. Aflatoxin entered into the human body through contaminated feed by using feed animal, such as crops, food, and so on, which caused of carcinogenic, teratogenic and other diseases to humans and animals. Therefore, it was great significance for accurate detection of aflatoxin content. Currently there was no detection technology of national standard for the aflatoxin of peanuts and peanut oil.In this study, improved high performance liquid chromatography(HPLC) was established for determination of aflatoxin in peanuts and peanut oil, samples pretreatment was optimized. Optimum chromatographic conditions were:This study to peanuts and aflatoxins in peanut oil extraction method of improvement, the result showed that Optimum extracting solution condition was acetonitrile and water(80:20 by volume), and recovery was the best. Mobile phase was obtained as acetonitrile and water; Flow rate was 0.5 m L/min; Excitation wavelength of fluorescence detection was Ex 360 nm, emission wavelength was Em 440 nm; Column temperature was 30℃; Injection volume was 25μL; Column of Waters PAHC18 was obtained(4.6mm × 150 mm, 5μm). Raw material storage of peanut oil production enterprises were by special sampling inspection for four areas(the pearl river delta, eastern of Guangdong, western of Guangdong and north of guangdong) of guangdong Province.The total sample of four regions involved with136 enterprises, including 136 batches of raw peanuts, the results showed that 16 batches of product failure, the overall pass rate of 88.24%. In failed enterprises, the pearl river delta region had three enterprises unqualified; In the western of Guangdong, the two companies were unqualified; In the northern part of Guangdong, eleven companies tested failed, which were the highest rates of failure. Research showed failure situations occured in the smaller companies. Another Zhaoqing, Yunfu, Yangjiang, Zhanjiang, Maoming and other five regions of Guangdong Province, a total of 200 small workshop in western Guangdong refined peanut oil sample survey, 200 batches of samples were detected in 112 batches of qualified, unqualified aflatoxin B1 88 batches of samples failed detection rate of 44.00%. |
