Coastal seawater,marine wastewater(ballast water,washing water,desulfurization waste water)and other waste water containing a higher concentration of ammonia and petroleum alkanes.The biological purification of such waste water,requires a high salt adaptability,high efficiency and degrading the prosperity of ammonia nitrogen and petroleum alkane simultaneously.In this paper,the use of anti-resistence additives to help the compound enzyme preparation of waste water purification technology,simulated ship wastewater denitrification and degradation of petroleum alkane.In this paper,a strain H02 capable of synthesizing Ectoine was screened from the salt pond in a salt field of Dalian.The strain H02 was identified by morphological identification and 16S rDNA as Halomonas strain,named Halomonas sp.H02.The synthesis of Halomonas sp.H02 was identified by high performance liquid chromatography and infrared spectroscopy.The results showed that it was Ectoine.The growth and Ectoine fermentation characteristics of Halomonas sp.H02 were investigated,and the optimum conditions were as follows:Halomonas sp.H02 was a non-secreting strain,and the optimum fermentation medium was MG modified medium,the optimum carbon source was sodium glutamate,the optimum salt concentration is 90 g/L.The factors affecting the synthesis of Ectoine were investigated by single factor experiment.The optimum Ectoine fermentation medium was obtained by response surface methodology.Ectoine was prepared by down-shock method.The average preparation rate was 94.0%.The optimum conditions for the determination of horseradish peroxidase(HRP)and laccase which commonly used in sewage treatment,and the protective effect of Ectoine on the two enzymes in extreme environment was investigated.The results showed that the optimal temperature of HRP was 30 ?,the optimum pH was 6,and the activity of HRP was significantly inhibited by NaCl concentration,Laccase was sensitive to salt,the optimum temperature was 40 ?,the optimum pH was 4.Ectoine has a significant effect on the protection of enzyme activity at high salt,extreme temperature,and extreme pH.The compound enzyme preparation was prepared by mixing HRP,laccase and other cells with biodegradable oil and denitrification,and adding Ectoine as anti-resistance assistant.The results showed that the oil degradation rate of up to 52.5%,and the denitrification rate was 44.2%.
