Polygonatum cyrtonema Hua(PCH),as a traditional medicinal and edible herb,has abundant bioactive components,such as flavonoids,polysaccharides,sanponins and amino acids,etc.,and anti-virus,anti-tumor and antihypertension properties.At present,PCH has been applied in cardiovascular disease prevention,cancer treatment and immunoregulation areas.Meanwhile,PCH also has antibacterial,moist and antioxidant effects and thus is pro mising to be applied in moisturizing,whitening and wrinkling cosmetics.In this thesis,based on solvent method and ethanol precipitation method,flavonoids and polysaccharides were extracted from PCH.On one hand,structures and total antioxidant capacity of the extracted active ingredients were analyzed and studied by different measurement methods and oxygen free radical elimination experiments,which provided a theoretical foundation for applications in antioxidant cosmetics.Major results of this thesis are as follows:1.The rhizome of PCH,as raw material,was extracted,the temperature,ethanol concentration,material ratio and extracting time of which were explored to find optimum total flavonoids extraction efficiency.The optimal extracting conditions of the total flavonoids were determined through orthogonal test: 70 ℃,90% ethanol,1:15 material ratio and 1 h.The extraction amount of total flavonoids was 0.211 mg/g under above-mentioned conditions.Polysaccharide of the extracting solution was precipitated through using 95% ethanol solution in which the 10-fold solvent was determined to be the optimal precipitation volume.The extraction amount of total polysaccharides was 150.9 mg/g.2.Qualitative and quantitative analyses of the extracted flavonoids were performed by high-pressure liquid chromatography(HPLC).The main components were determined to be rutin,quercetin and diosgenin,and their contents were 0.036,0.132 and 0.255 mg/g,respectively.The structures of the polysaccharides of extract were analyzed by gas chromatography-mass spectroscopy(GC-MS)and infrared spectroscopy.These results showed that PCH contained pentosans and long-chain polysaccharides with more than 16 carbons.Furthermore,the polysaccharides existed as acidic polysaccharide structure.3.The antioxidant activity of the extracts was evaluated by a model test of diphenyl picryl hydrazinyl radical(DPPH).The elimination rate of oxygen free radicals of the extracts was >90% when 0.8 mL 0.1 mg/mL extracts were used,which was far higher than commercial antioxidant(2,6-di-tert-butyl-4-methylphenol).Moreover,the cosmetic formulations were designed and prepared through adding extracts as natural antioxidant ingredients,showing that the extracts had good stability and compatibility in the formulations.
