Objective: Polygonatum cyrtonema Hua in Jiuhua mountain is famous in polygonatum sibiricum for its high quality,it comes from Liliaceae plant Polygonatum cyrtonema.Polygonatum cyrtonema Hua in Jiuhua mountain is mainly produced in the area of Mount Jiuhua,Anhui province.Due to excessive excavation,the resource variety is endangered,There are often other origin of polygonatum sibiricum counterfeit Polygonatum cyrtonema Hua on the market,resulting in domestic market chaos,resulting in uneven quality of origin of polygonatum sibiricum.Therefore,in this experiment,we used Polygonatum cyrtonema Hua in Jiuhua mountain,Polygonatum cyrtonema Hua and Polygonati Rhizoma Red.Polygonatum kingianum Coll.et Hemsl.as materials to identify the differences of polygonatum sibiricum from different bases and to explore the genuineness of polygonatumsibiricum.Methods: Studyed the genetic background of polygonatum sibiricum from different sources by ISSR molecular marker,and determined the content of polysaccharide in dried and fresh polygonatum sibiricum by sulfuric acid-anthrone method.Used the HPLC fingerprint to establish the characteristic chromatogram of Polygonatumcyrtonema Hua in Jiuhua mountain.Results: 1 Analyzed the genetic background of different varieties of polygonatum sibiricum by ISSR molecular marker technique,Screened 10 polymorphic primers out from 100 random primers,and amplified 107 DNA bands,among which 105 were polymorphic bands.From the results of cluster analysis,it shows that Polygonatum cyrtonema Hua is not related to the other two kinds of Polygonati Rhizoma,the intraspecific genetic diversity of Polygonatum cyrtonema Hua is related to geographical environment,which is consistent with the theory of genuine medicinal materials.2 Compared the content of polysaccharides in different varieties of Polygonati Rhizoma.The results showed that Polygonatum cyrtonema Hua ’s polysaccharide content was the highest among the Polygonati Rhizoma,The content of polysaccharides in fresh and dried polygonatum sibiricum from the same source is significantly higher than that in dried polygonatum sibiricum,As the main active component of polygonatum sibiricum,it is necessary to study the change of the component of polygonatum sibiricum during drying and its effect on pharmacological activity,so as to provides the basis for the more reasonable and effective development of polygonatumsibiricumpolysaccharides.3 The characteristic fingerprint of Polygonati Rhizoma was established and 11 common peaks(11 Diosgenin Peak)were demarcated.The similarity analysis showed that the fingerprints of different varieties of Polygonati Rhizoma from different habitats were obviously different.It is helpful for the identification and quality control of Polygonati Rhizoma.Conclusion: The method and data analysis can provide scientific basis for improving quality standard of Polygonatum cyrtonema and authenticity of Jiuhua Mountain polygonatu.It can also provide scientific basis for clarifying the genuineness of polygonatumsibiricum.
