Objective:By establishing the animal models of total knee arthroplasty,we will observe the variation of early preoperative blood loss with tranexamic acid,and study the effect of tranexamic acid on hidden blood loss and its influence on deep venous thrombosis.Methods:We designed to complete the rabbit knee prosthesis ourselves,20 adult New Zealand rabbits were divided into two groups at random,each group of 10,all were conducted with the same side TKA,in experimental group rabbits were dropped 0.5 g 100 ml of tranexamic acid in preoperative 15 points,the control group was given 100 ml of 0.9%sodium chloride at the same time,all experimental animals were drawn in ear marginal vein blood in preoperative day 12-14 h,postoperative 3 h、6 h、24 h、48h、72h and the 7th day respectively,we repeatedly checked blood routine examination and blood coagulation function,determined the content of plasma D-D dimmer,meanwhile,we recorded the operation time and bleeding amount of calculation,have been Unified statistical analysis and conducted lower limb venous ultrasound post-operation.Results:There is no significant difference in the experimental group and control group on preoperative weight,Hb,Hct and D-D dimer(p>0.05).The difference of operative time and intraoperative explicit differences in blood loss has a significant meaning(p<0.05),by compared,the values of Hb and Hct after 24h have obvious differences,and no differences between two groups on D-D dimmer(p>0.05),they have no obvious correlation with the change of Hct,、Hb after postoperative 72 h、the 7th day and the variation of perioperative D-D dimer.There is no significant postoperative lower limb venous ultrasound thrombus.Conclusion:By means of establishing animal models of artificial knee joint replacement in this article,we observed the variation of perioperative blood loss after using tranexamic acid,it was shown that tranexamic acid could significantly reduce intraoperative dominant blood loss,shorten operating time,and could significantly decrease the hidden blood loss after the first day which resulted from fibrinolysis with surgical stress,no obvious effect on another unless other mechanisms.Meanwhile,it reserved safe and reliable,and not increase the risk of perioperative blood clots.This study provided the theory basis for clinical use of tranexamic acid,serving for exploring the mechanism of hidden blood loss. |
