The Animal Experiments Research About Fuzheng Tongluo Soup On Chronic Fatigue Syndrome

Objective:In the present study,we intended to observed the effects and mechanism of Fuzheng Tongluo soup on anti-stress normal mice and lipid peroxides and neurotransmitters in model mice of fatigue syndrome.Methods:(1)Mice were randomly divided into normal control group,model group,large dose group,middle dose group,small dose group.Different doses of drugs were given to mice for 14 days.The period of the experiment,the mice were drank and feed by themselves.After 1 hour in last administration,normotensive hypoxia tolerance test was conducted.The other mice,packet and administration were ditto.Swimming test,cold tolerance test and thermostable test were performed separately.(2)In addition,50 mice were randomly divided into normal control group,model group,large dose group,middle dose group,small dose group.Except normal control group,Dexamethasone(DSMS)was injected 14 days to cause the immunosuppression of mice.After administration next day,all of the mice were killed off and took out the thymus and weighed it,while thymus index was calculated.Taking out of the another thymus histopathology and conducting dyeing by HE to observed the effects of Fuzheng Tongluo soup on thymus index and pathological changed.(3)In addition,50 mice were randomly divided normal control group,model group,large dose group,middle dose group,small dose group.Different doses of drug were given to mice for 20 days.Suitability suspension and swimming 10 min at first 3 days,after this each 4 days for increasing suitability suspension and swimming 10 min amount of circulation.The model was performed in this way until end of the experiment.After 30 min in last administration,other group swam 15 min and had a rest 30 min except for normal control group,Picking out the eyeball and collecting blood,livers,while taking out the hypothalamus and pituitary detect levels of SOD,MDA,glycogen,BUN,GSH-PX and 5-HT,DA content.Result:(1)Compare with normal control group,the normotensive hypoxia tolerance time,swimming time and the thermostable time under the above conditions were both pro-longed(P<0.01 or P<0.05).The large dose group,middle dose group can improve cold tolerance time(P<0.01).Compare with model group,The large dose group,middle dose group can improve swimming time and thermostable time,while small dose group can improve thermostable time only(P<0.05).(2)Fuzheng Tongluo soup can improve the shrinking thymus and increase the thymus index(P<0.01 or P<0.05).(3)Compare with modle control group,Fuzheng Tongluo soup can significantly changed the level of glycogen,BUN,SOD,MDA,GSH-PX and 5-HT,DA(P<0.01 or P<0.05).Conclusion:(1)Fuzheng Tongluo soup can enhance the ability of anti-stress of mice.(2)Fuzheng Tongluo soup can increase the thymus index,improve thymus atrophy status,enhance the amount of thymus cortical cells and improve immune function.(3)Fuzheng Tongluo soup can enhance the ability of anti-fatigue of mice.
