| Objective:Using the ploycystisc ovary(PCO)rats fed by letrozole as the study object,to research RANTES and Mfn2 expression in uterus of rats with PCO.Then these rats were treated with add-subtraction bushenyangjing decoction to further observe the changes of RANTES and Mfn2 expression in uterus of rats with PCO and discuss the mechanism of add-subtraction bushenyangjing decoction which is used to treat PCOS.Methods:64 SD female rats,whose age is 5 weeks.After adaptively feeding one week,these rats were randomly divided into solvent control group(n=16)and PCO model group(n=48)which were fed 21 days by letozole.Through observing vaginal secretion smear and hormone detection to determine the model is successful.Then the PCO model rats were randomly divided into model control group,high,medium and low dosage of add-subtraction bushenyangjing decoction group,and clomifene citrate group.After treating and kiling the animals,using the immu-nohistochemistry streptavidin peroxidase conjugated method to examine the experssion of RANTES and Mfn2 in the uterus,and then,using the microscope to observe and graded by double blind method to compare the differences among these groups.Results:RANTES and Mfn2 express in the cells of uterine epithelium in all the groups.1.Compared to the solvent control group,the expression of RANTES in model group was much greater(P<0.01);Compared to the model control group,the expression of RANTES in high,medium and low dosage of add-subtraction bushenyangjing decoction group were much lower(P<0.01);Compared to the model group,the expression of RANTES in the clomifene citrate group was much lower(P<0.05);Compared to the clomifene citrate group,the expression of RANTES in the high dosage of add-subtraction bushenyangjing decoction group was much lower(P<0.01).Although the expression of RANTES in the medium and low dosage of add-subtraction bushenyangjing decoction group were much lower when compared to the clomifene citrate group,it did not have the statistical significance(P>0.05).2.Compared to the solvent control group,the expression of Mfn2 in model group was much lower(P<0.01);Compared to the model control group,the expression of Mfn2 in the high,medium and low dosage of add-subtraction bushenyangjing decoction were much greater(P<0.01);the clomifene citrate group was much greater when compared to the model group(P<0.05);Compared to the clomifene citrate group,the expression of Mfn2 in the high dosage of add-subtraction bushenyangjing decoction group was much greater(P<0.01).Although the expression of Mfn2 in the medium and low dosage of add-subtraction bushenyangjing decoction group were much greater when compared to the clomifene citrate group,it did not have the statistical significance(P>0.05);3.By analyzing,the relationship of RANTES and Mfn2 is markedly negative correlation(r=-0.634,P<0.01).Conclusion:1.Compared to the solvent control group,the expression of RANTES in the model control group was much greater,while the expression of Mfn2 was much lower,we infer that RANTES and Mfn2 can influence the internal environment of uterus by insulin resistence which can influence the fetation;2.In the model control group,the add-subtraction bushenyangjing decoction can made RANTES down-regulated expression,while Mfn2 up-regulated expression,we infer that the add-subtraction bushenyangjing decoction can improve the internal environment of uterus and increase the rate of pregnancy;3.Compared to the medium and low dosage of add-subtraction bushenyangjing decoction group,the change of RANTES and Mfn2 expression in the uterus of the high dosage of add-subtraction bushenyangjing decoction group is significant,we infer that treating PCOS with add-subtraction bushenyangjing decoction may have a dose-dependent manner;4.In model group,the expression of RANTES is up-regulation,while,the expression of Mfn2 is down-regulation,negative correlation between them,we infer that there is a common signal path with them,by which they can interact on each other and influence the internal environment of uterus. |
