Objective:To observe the influence of bushenyangjing decoction on Bax,Bcl-2 and Fas expression in ovary of rats with PCO and investigate the mechanism of bushenyangjing decoction treating the abnormal ovulating of PCO.Methods:50 SD female rats,23 days age,were divided randomly into model group including 40 rats and normal control group including 10 rats.Then the rat model with PCO was induced by DHEA,40 model rats were divided randomly into model control group,clomifene citrate group,high dosage of bushenyangjing decoction group,low dosage of bushenyangjing decoction group.After treatment and killing the animal,the expressions of Bax,Bcl-2 and Fas proteins with immunohistochemistry streptavidin peroxidase conjugated method were examined and observed by optical density value with microscope and analyzed by computer imaging technique software named Image-Pro plus 6.0. Results:1.Through imaging and statistics analyzing,Bax,Fas and Bcl-2 protein expressed in follicles of polycystic ovary in all the groups including model, normal control group,clomifene citrate group,high dosage of bushenyangjing decoction and low dosage of bushenyangjing decoction group. The OD expressions of Bax and Fas protein in model group were much greater than that in normal control group, but Bcl-2 protein was much smaller (P<0.05);The OD expressions of Bax and Fas protein in all the medicine groups including the high dosage of bushenyangjing decoction, low dosage of bushenyangjing decoction and clomifene citrate group were much smaller than that in model group, but Bcl-2 protein was much greater (P<0.05); The OD expressions of Bax and Fas protein in the high dosage of bushenyangjing decoction group were much smaller than that in the clomifene citrate group, but Bcl-2 protein was much greater (P<0.05), the low dosage of bushenyangjing decoction was little greater than that in the clomifene citrate group, but Bcl-2 protein was little smaller (P<0.05). The OD expressions of Bax and Fas protein in the high dosage of bushenyangjing decoction group were much smaller than that in the low dosage group, but Bcl-2 protein was much greater (P<0.05).2.The expressions of Bax,Fas two types of apoptosis induced protein and Bcl-2 of apoptosis inhibiting protein in all the groups including model, normal control group,clomifene citrate group,high dosage of bushenyangjing decoction and low dosage of bushenyangjing decoction group were negatively correlated(P<0.05), but Bax and Fas two types of apoptosis induced protein were no obvious correlation in all the groups (P>0.05). Conclusion:The rat model with PCO was induced by DHEA,then we affirm it was successful by exfoliative cytoscopy of vagina, detection of sex hormone in serum and ovarian histopathological observation.The apoptosis of ovary granulosa cells increased obviously in model group, it is a extremely important causation of PCO that the high expressions of Bax, Fas and the low expression of Bcl-2 at course of follicles growth.Bushenyangjing decoction can increase the expression level of Bcl-2 and reduce the expression levels of Bax and Fas in follicles granulosa cells of polycystic ovary in the model group,It can inhibit the apoptosis ovary granulosa cells by regularing the blance of Bcl-2/Bax system and decreasing the expression level of Fas in follicles for the purpose of treating PCO.
