Objective: To verify the effectiveness of acupuncture for diarrhea,this paper by copying diarrhea rabbit model to rabbit intestinal propulsive rate index,and as a starting point, compare the difference between acupoints and synergies;And on this basis, by observing the morphology of intestinal mucosa of rabbits before and after acupuncture, the SP intestinal tissue, changes in the expression of VIP, and explore the mechanism of acupuncture intervention in regulating the movement of intestinal diarrhea rabbit model, to provide the experimental basis for Meridian-Viscera Doctrine.Methods: Choose 60 healthy New Zealand white rabbits who's body quality for 2~2.5 kg, divide into several groups in random: normal group, model group,tianshu group dachangshu group quchi group shangjuxu group, Tianshu + Dachangshu group,Quchi + Shangjuxu group,those groups of rabbits are ten.The diarrhea models of rabbits were made by filling the decoction of Folium sennae method, the Tianshu group, Dachangshu group Quchi group, Shangjuxu group take bilateral acupoint acupuncture, Tianshu + Dachangshu group,Quchi + Shangjuxu group take unilateral acupuncture points, Acupuncture needle into, hand-held needle operation to 60 times per minute frequency, twisting 1min,retaining needl 20 min, every 10 min line pin 1, and continuous 3days.the normal group treat normally,no modeling, no acupuncture,model group was only carried out without acupuncture treatment. Fasting semi-solid carbon powder paste 20ml/kg after the last treatment. 10 minutes later, the rabbits were killed by gas embolism.Open,remove the small intestine quickly,From the pylorus of small intestine in turn after peeling off the tile on a white sheet of paper, measurements of the pyloric back to the ileocecal valve at length and recording, measuring to the pylorus appeared for the first time and record the distance semi solid charcoal nutritious paste. After distance accounted for the percentage of the former distance as small intestinal propulsion rate.And take the corresponding position 1 cm long intestinal tissue placed in 4% neutral formalin fixed for immunohistochemistry.Results:(1) Acupuncture single points on intestinal propulsion rate impact : Compared with the normal group,the model group( P < 0.01), the difference is statistical significant. In addition to acupuncture Shangjuxu(ST37) group has no significant effect on the inhibition of intestinal propulsion rate of diarrhea rabbits, the other groups all significantly lower the intestinal propulsive rate;compared with the model group, Tianshu(ST25) group( P < 0.01), Dachangshu(BL25) group(P < 0.01), Qu Chi(LI11) group( P < 0.01), the differences have statistical significance, which Tianshu(ST25) group on rabbit intestinal propulsive rate decreased most significantly, Dachangshu(BL25) group followed.(2) Acupuncture single points and the corresponding points set on intestinal propulsive rate impact : Compared with the model group, Qu Chi(LI11) + Shangjuxu(ST37) group(P<0.01),the intestinal propulsion rate of diarrhea rabbits is significantly reduced,Compared with Qu Chi(LI11) group, Qu Chi(LI11) + Shangjuxu(ST37) group(P>0.05), the difference has no statistical significance; Compared with the model group, Tianshu(ST25) + Dachangshu(BL25) group significantly lower the intestinal propulsion rate of diarrhea rabbits; And compared with the Tianshu(ST25) group and Dachangshu(BL25) group, Tianshu(ST25) + Dachangshu(BL25) group(P<0.05), the differences have significant statistical significance.(3) Intestinal tissue morphological changes: light microscope, the normal structure intact intestinal mucosa villi intact, no obvious inflammatory cell infiltration; structure is incomplete model group, small intestine, wall thinning, mucosal swelling in different degrees of congestion, inflammatory cells invasion, microvilli atrophy, loss, defect structure of intercellular connections phenomenon; significant improvement in the treatment group pathological intestinal mucosa, mucosa only mild swelling, clear structure, almost no hair loss.(4) Intestinal tissue VIP, SP expression: Compared with normal group, model group, intestinal tissue VIP, SP immune expression level was significantly increased(P <0.01), compared with the model group, Tianshu, Dachangshu small intestine tissue VIP, SP immunity a significant reduction in the number of positive cells(P <0.01).Conclusion:(1)Acupuncture can inhibit diarrhea condition of rabbits small intestine movement, and the effect of the acupuncture play with acupuncture is closely related to the specificity of acupoints. Acupuncture can used in the treatment of diarrhea.(2)Acupuncture treatment of diarrhea assocoated with acupuncture of acupoints compatibility synergy, through acupoints compatibility synergies, acupuncture can better play the role of the functions of regulating the zang-fu organs.(3)Acupuncture can be used in the treatment of diarrhea may be related to improving the pathological damage of the damaged small intestinal mucosa.(4)The mechanism of acupuncture treating diarrhea may be realized by down regulating the expression level of VIP and SP in small intestinal tissue.
