Objective : To explore the effect of Danshentongluojiedu Decoction on the TLR4/ NF-κB signal pathway and downstream factor in acute myocardial infarction(AMI) rats by Bone marrow stem cell transplantation.Methods : The whole bone marrow adherent method was adopted for BMSCs separation and culture. The AMI model was established by closing the left anterior descending coronary artery of SD rats. After the success, SD rats were randomly divided into SH(sham group), AMI group(acute myocardial infarction group), BMSCs(BMSCs transplantation group), DSTLJD group(Danshentongluojiedu group), DSTLJD + BMSCs group(Danshentongluojieducombined with BMSCs transplantation group), 10 in each group. BMSCs cell suspension injected directly into the edge of the myocardial tissue infarction area; Chinese medicine or physiological saline were administered by gavage.4 weeks later, by using enzyme-linked immunosorbent(ELISA) measure each group rats serum TNF–α,IL–6, MCP-1,sICAM-1 content. Immunohistochemical method to observe the myocardial NF-κB protein expression, the expression TLR-4 was measured by western blotting,and HE staining to observe the myocardial tissue pathological changes.Result:1.Compared with the acute myocardial infarction group, BMSCs group, DSTLJD group, DSTLJD + BMSCs group rats serum level of TNF-α,IL – 6,MCP-1,sICAM-1 were significantly reduced, the difference has statistically significant(P<0.01, P<0.05); Among them, DSTLJD group was better than that of BMSCs transplantation group,DSTLJD + BMSCs group was better than that of pure stem cell transplantation, and simple DSTLJD group, the difference has statistically significant(P<0.01, P<0.05).2. Compared with the acute myocardial infarction group, BMSCs group, DSTLJD group, DSTLJD + BMSCs group rats myocardial injury was significantly improved,myocardial nuclear factor NF-κB protein expression and myocardial TLR-4 protein expression were significantly reduced, the difference has statistically significant(P<0.01, P<0.05); As well as, DSTLJD group was better than that of BMSCs transplantation group,DSTLJD + BMSCs group was better than that of pure stem cell transplantation, and simple DSTLJD group, the difference has statistically significant(P<0.01, P<0.05).Conclusion:1.The protective effect of Danshentongluojiedu Decoction on the rats with acute myocardial infarction may be related to the inhibition of inflammatory reaction,that’s mechanism may be related to the regulation of TLR4/NF-κB Signal transduction pathways TNF-α, IL-6, sICAM-1, and MCP-1 of downstream.2. The protective effect of Danshentongluojiedu Decoction combined with BMSC on rats with acute myocardial infarction were better than simple Danshentongluojiedu Decoction group and stem cell transplantation group,that’s mechanism may be related to the regulation of TLR4/NF-κB’s inflammation signal.
