Objective: 1.Stachys japonica Miq. is an perennial herb from Labiatae Stachys. This paper aimed to study the chemical compositions of the extract of Stachys japonica Miq.2.And to investigate the immunoregulation action of the herb in an experimental animal model of humoral immunity, cellular immunity, non-specific immunity activity of the ethanol extract of Stachys japonica Miq. 3. At the same time, this experiment determine antioxidant activity of different polar parts of the ethanol extract S. japonica(SJEE) and the component analysis, to track S. japonica(SJEE). antioxidant activity efficacy material base.Methods: 1. Adopt the method of qualitative test, test S. japonica. contains the main chemical composition. 2. Humoral immunity was evaluated using serum hemolysin.Cellular immunity was evaluated using DNFB-induced delayed-type hypersensitivity reaction. Non-specific immune function was evaluated by using immune organ index and carbon clearance assay. The immunosuppressive mouse model was established by injection of cyclophosphamide(80mg/kg) into the abdominal cavities of normal mice.SJEE was administered by gavage at low dose, medium dose, and high dose of 1, 2, and4g/kg, respectively. 3. In this study, We using the bioassay method to analyze of antioxidant activity of petroleum ether extract, chloroform extract, ethyl acetate extract,n-butanol extract and aqueous extract. The antioxidant activities of SJEE were determined by 2,2-diphenyl-1-picrylhydrazyl radical scavenging assay, superoxide radical assay, hydroxyl radical assay and reducing power assay. Ascorbic acid and BHT were used as positive control.Results: 1. The main chemical composition of S. japonica contains flavonoids,polyphenols, tannins, alkaloids, saponins, polysaccharides, steroids, terpenoids and other compounds. 2. The SJEE(2 and 4g/kg body weight) significantly improved the humoral immune response, cellular immune response, immune organ index, and phagocytic activity of immunosuppressive mice. 3. The SJEE showed remarkable antioxidant activity. Among the five extract, the ethyl acetate extract showed the strongest antioxidant activity,Flavonoids and polyphenols contained in ethyl acetate polar than other parts of high polarity.Conclusion: S. japonica main chemical composition contains flavonoids, polyphenols,tannins, alkaloids, saponins, polysaccharides, steroids, terpenoids and other compounds.S. japonica can significantly improve immune function in immunocompromised mice and has a significant role as an antioxidant.
