Objective: To investigate the expression of LKB1,AMPK and mTOR in Laryngeal squamous Cell Carcinoma,and explore the role of LKB1,AMPK and mTOR in the occurrence and development of laryngeal cancer by analyzing their expression levels and their relationship with clinicopathologic factors,thereby provide theoretical basis for the early diagnosis and targeted treatment of Laryngeal squamous Cell Carcinoma.Methods: A total of 42 cases laryngeal squamous cell carcinoma tissue samples were collected as experimental group,at the same time,collected the 42 cases laryngeal carcinoma adjacent tissues accordingly as comparison group.The expressions of LKB1,PK and mTOR in tissues of the two groups were measured by streptavidin peroxidase(SP)immunohistochemical method,and the relationship between the expressions of LKB1,AMPK,mTOR with clinicopathological parameters was analysed.The correlation of LKB1,AMPK,mTOR with laryngeal squamous cell carcinoma was analyzed by Spearman rank analysis.Results:1 the expression of LKB1: The positive expression rate of LKB1,in laryngeal carcinoma tissue,adjacent normal tissue,high differentiation laryngeal carcinoma tissue,middle and low differentiation laryngeal carcinoma tissue,with lmph node metastasis group,without lmph node metastasis group,Ⅰ ~ Ⅱ stage laryngeal carcinoma group,Ⅲ ~ Ⅳ stage laryngeal carcinoma group,were 66.7%,85.7%,84.2%,52.2%,78.6%,42.9%,81.8%,50.0% respectively.The expression difference of the four groups were statistieally signifieant(P<0.05),suggested that the expression of LKB1 in laryngeal carcinoma tissue was lower than in adjacent normal tissue,and it may related to the degree of pathological differentiation,lymph node metastasis and clinical stage of laryngeal squamous cell careinoma.The positive expression rate of LKB1,in laryngeal carcinoma patients less than or equal to 60 years of age,over 60 years of age,supraglottic carcinoma group,glottic carcinoma group,somoking patients with smoking less than or equal to 20 a day,somoking patients with smoking over 20 a day,were 64.7%,68.0%,69.2%,65.5%,66.7%,66.7% respectively.The expression difference of the above three groups were no statistieally signifieant(P>0.05).This consequence suggested that the expression of LKB1 in laryngeal carcinoma tissue is not related to patient age,primary tumor position and smoking.2 the expression of AMPK: The positive expression rate of AMPK,in laryngeal carcinoma tissue,adjacent normal tissue,high differentiation laryngeal carcinoma tissue,middle and low differentiation laryngeal carcinoma tissue,with lmph node metastasis group,without lmph node metastasis group,Ⅰ ~ Ⅱ stage laryngeal carcinoma group,Ⅲ ~ Ⅳ stage laryngeal carcinoma group,were 45.2%,78.6%,68.4%,26.1%,57.1%,21.4%,68.2%,20.0% respectively.The expression difference of the four groups were statistieally signifieant(P<0.05),suggested that the expression of AMPK in laryngeal carcinoma tissue may related to the degree of carcinoma pathological differentiation,lymph node metastasis and clinical stage.The positive expression rate of AMPK,in laryngeal carcinoma patients less than or equal to 60 years of age,over 60 years of age,supraglottic carcinoma group,glottic carcinoma group,somoking patients with smoking less than or equal to 20 a day,somoking patients with smoking over 20 a day,were 52.9%,40.0%,53.8%,41.4%,48.1%,40.0% respectively.The expression difference of the above three groups were no statistieally signifieant(P>0.05).This consequence suggested that the expression of AMPK in laryngeal carcinoma tissue is not related to patient age,primary tumor position and smoking.3 the expression of mTOR: The positive expression rate of mTOR,in laryngeal carcinoma tissue,adjacent normal tissue,high differentiation laryngeal carcinoma tissue,middle and low differentiation laryngeal carcinoma tissue,with lmph node metastasis group,without lmph node metastasis group,Ⅰ ~ Ⅱ stage laryngeal carcinoma group,Ⅲ ~ Ⅳ stage laryngeal carcinoma group,were 69.0%,28.6%,42.1%,91.3%,57.1%,92.9%,54.5%,85.0% respectively.The expression difference of the four groups were statistieally signifieant(P<0.05),suggested that the expression of mTOR in laryngeal carcinoma tissue may related to the degree of carcinoma pathological differentiation,lymph node metastasis and clinical stage.The positive expression rate of mTOR,in laryngeal carcinoma patients less than or equal to 60 years of age,over 60 years of age,supraglottic carcinoma group,glottic carcinoma group,somoking patients with smoking less than or equal to 20 a day,somoking patients with smoking over 20 a day,were 64.7%,72.0%,69.2%,69.0%,66.7%,73.3% respectively.The expression difference of the above three groups were no statistieally signifieant(P>0.05).This consequence suggested that the expression of mTOR in laryngeal carcinoma tissue is not related to patient age,primary tumor position and smoking.4 the correlation among LKB1,AMPK and mTOR expression level: The date analyzed by Spearman’s Rank Correlation Analysis showed that the expression of LKB1 was positive related to the expression of AMPK(rs=0.77,P=0.000).The expression of LKB1,AMPK was respectively negative related to mTOR.(rs=-0.94,P=0.000;rs=-0.89,P=0.000).Conclusion:1 The expression of LKB1 and AMPK in laryngeal carcinoma tissue were lower than that in adjacent normal tissue,while the expression of mTOR was higher.The expression of LKB1,AMPK,mTOR were related to the degree of carcinoma pathological differentiation,lymph node metastasis and clinical stage,regardless of patient age,primary tumor position and smoking.2 LKB1 expression level was positive related to the AMPK expression level,while LKB1,AMPK was all negative related to mTOR.LKB1 could active AMPK,then the activated AMPK negatively regulated the expression of mTOR.Therefore,the carcinoma occur when the negative regulation was blocked due to the activity of LKB1 and AMPK were decrease.3 The expression of LKB1 AMKP and mTOR in laryngeal carcinoma tissue is not related to patient age and smoking.But some studies prove that patient age and smoking has certain correlation with the incidence of laryngeal carcinoma development,this study may be due to less sample cases,need to be studied further.4 The study provide a theoretical basis for early diagnosis of Laryngeal squamous Cell Carcinoma.The developing of LKB1 and AMPK agonist,as well as mTOR inhibitors may become a new methods to treatment Laryngeal squamous Cell Carcinoma.
