Objective: To investigate the effect of lian-huang zi compound on the xenografted tumor of the human bladder cancer T24 cell line on nude mice and to discuss its mechanism.Methods: The xenografted tumor of bladder cancer T24 cell model was established in nude mice which were then randomized to receive treatment with Physiological saline group control group(control group, 0.9%NS), low concentration of lian huang zi compound group(2.5ml/kg/day)and high concentration group(5ml/kg/day),that were filled the stomach medicine every other day and for seven times. All mice were measuring tumor volume and sacrificed after the next day of last administration for performing following experiments,measuring the mice weight and the tumor weight. The serum of xenografted tumor mice were collected and analyzed by ELlSA to the expression of TNF-?, GM-CSF, IFN-?.The expression of apoptosis associated protein bcl-2 and bax, associated antioncogene PENT by immunocytochemistry.Results: The xenografted tumor of the human bladder cancer T24 cell line on nude mice were suppressed significantly by the lian-huang zi compound. That's More obvious than its low concentration of lian huang zi compound group and high concentration group, which have the tumor inhibition rate ratios of 52.14%, 66.67% respectively. The ELISA results showed that lian-huang zi compound. can reduce the expression of GM-CSF?IFN-? in xenografted tumor serum, increase the expressiom of TNF-?. The expression of apoptosis associated protein bcl-2 obviously reduced, but the bax protein and antioncogene pten was significantly increased. The tumor inhibition rate and apoptosis rate had the increasing tendency, the expression of apoptosis associated protein bcl-2 were obviously reduced, but the bax protein and pten protein was significantly increased with the increasing concentrations of its recipe.Conclusion: The lian-huang zi compound had obviously suppressed the xenografted tumor of the human bladder cancer T24 cell line,which may be related partly through the Induction of apoptosis, its up-regulation of BaX and PTEN down- regulation of Bcl2.
