Objective: In this study,the expression of different serum proteins extracted from ST segment of elderly patients with coronary elevation acute myocardial infarction and normal elderly people was analyzed and explored by iTRAQ technology and bioinformatics analysis.This study can further research the occurrence and development of warning proteins in elderly myocardial infarction patients and provided the theoretical and experimental basic data for finding the risk factors or potential therapeutic targets.Methods: Quantitative proteomic iTRAQ method was applied to analyze the composition of serum proteins.10 serum proteins samples were extracted from elderly patients with ST-segment elevation acute myocardial infarction;meanwhile,the normal serum proteins from healthy elderly people acted as the control.After remove the peak protein of obtained serum proteins,the residual proteins were prepared as 12% SDS polyacrylamide gel.Subsequently,the gel was digested by trypsin and enzymolysis peptides were labeled by iTRAQ reagent.Finally,the labeled peptides were analyzed by LC-MS/MS mass spectrum and bioinformatics analysis methods.Results: This study was the first time to analyze the serum proteins by using the quantitative proteomic iTRAQ technology.95 kinds of proteins,which extracted from elderly patients with ST-segment elevation acute myocardial infarction,showed the difference as compared to the normal serum proteins.Among them,43 kinds of proteins displayed the remarkable increased tendency,and 52 of them exhibited the extremely decreased tendency.All these different proteins involved 62 kinds of biological processes,18 kinds of cellular components and 13 kinds of molecular pathways.The KEGG analysis results showed that 4 pathways including the complement and coagulation cascade pathway,systemic lupus erythematosus path,pathogenicity E.coli infection pathways displayed the protein-rich phenomenon.All these signaling pathways might play an important role in the formation process of elderly patients with acute myocardial infarction.Conclusion: In this study,quantitative proteomic iTRAQ technology was firstly used to analyze the serum proteins extracted from elderly patients with acute myocardial infarction.Comparing to the normal serum proteins,95 kinds of different proteins were identified and analyzed.To analyze the different proteins to the elderly patients with acute myocardial infarction by using bioinformatics analysis method will be beneficial to monitor the level of FIG,PLG,C6,and C4 B in serum and the specific treatment to the FIG,besides,will be good for forecasting the acute STEMI in elderly people,illustration of pathogenesis and drug targeted therapy.
