Background In recent years,the incidence of breast cancer continues to rise,seriously endangering the health of women.In some cities of China,the incidence of breast cancer has been located in the first place of female malignant tumors.Breast cancer has the characteristics of easy recurrence and metastasis.Therefore,screening suspicious cases to find early breast cancer patients have important significance.HPSE plays an important role in the recurrence or metastasis of breast cancer.This experiment,by polymerase chain reaction(RT-PCR)technique,detect heparanase mRNA expression in peripheral blood white cells of breast cancer patients before and after operation and the expression levels of healthy people and to explore the significance of heparanase mRNA in the review and survey of breast cancer patients.Objective To investigate the expression of HPSEmRNA in peripheral blood leukocytes and Tumor specimens and tissue adjacent to carcinoma of patients with breast cancer and the difference of the expression level after operation.Methods Collecting 45 cases of peripheral blood before and 9 days after operation of breast cancers from the general surgery department of First Affiliated Hospital of Xinxiang Medical College from 2015 June to October.All were female,aged 35-67 years old,the median age was 50.64± 0.88 years old,and also collect 10 morning fasting peripheral venous blood samples as the control group from the other patients with benign tumor of the breast.All cases had not been treated with radiotherapy or chemotherapy before operation.The clinical stage according to the TNM standard.Extracting peripheral venous blood 1ml of the patients,extracting total RNA according to the requirements.Amplificating heparanase mRNA through the application of RT-PCR,then after the agarose gel electrophoresis and photograph,determinating the band gray value.Application SPSS17.0 software for statistical analysis,measurement data was statisted by mean±standard deviation((?) ±s),Pairwise comparison was represented by Student t test.Count data comparison was represented by Chi square test.Inspection standard α=0.05.Results Heparanase mRNA in patients with breast cancer in the peripheral blood expression: after30 RT-PCR cycles,45 cases of breast cancer patients peripheral blood with 27 cases(60%)of the expression of heparanase mRNA,expression was 53.7%±4.7%,10 cases in the control group were all negative values,45 cases of breast cancer tissue heparan enzyme mRNA expression was 35 cases(77.8%),45 cases of tissue adjacent to carcinoma was for 11 cases(24.4%).Expression of heparanase mRNA has nothing to do with age;and tumor size,lymph node metastasis,tumor staging was related.The peripheral blood mRNA expression level of heparanase decreased significantly after surgery in patients with breast cancer(p<0.001).Conclusion Heparanase in breast cancer patients peripheral blood showed positive expression,its expression has nothing to do with the patient’s age,and tumor size,lymph node metastasis,neoplasm stage was related;the peripheral blood of heparanase mRNA expression level after surgery,in patients with breast cancer,decreased significantly.The peripheral blood of heparanase mRNA in breast carcinoma may be a good indicator of screening,evaluation postoperative follow-up and follow-up after operation.
