Background:Breast cancer is the most common malignant tumor which threatens women’s health and survival.Therefore,it is of geat importance to explore a marker,which has high specificity and high sensitivity,for early detection early diagnosis and early treatment of breast cancer.Accumulating evidence has shown that ubiquitin-conjugating enzyme 2 C(UBE2C)is highly expressed in breast cancer and it plays an important role in the occurrence and progression of breast cancer.It is expected to be an important marker of breast cancer in the future.However,the specific mechanism of UBE2 C in breast cancer and its relationship with the clinical pathological characteristics of breast cancer is still unclear.Futhermore,as a single marker for breast cancer,UBE2 C alone is still limited.In addition,P53 as an important tumor suppressor gene in a variety of tumors,including breast cancer,the mutation,loss of normal tumor suppressor role,making the tumor cell apoptosis decreased,growth increased.In the study of lung cancer,the expression of UBE2 C was positively correlated with the expression of mutant P53,And the cytological studies in glioma cells have found that UBE2 C may promote the development of tumor by reducing the P53 of wild-type tumor suppressor genes.Nevertheless,the correlation between UBE2 C and P53 in breast cancer and its clin ical significance have been rarely studied.Objective:To determine the expression of UBE2 C and p53 and explore their correlation in breast cancer tissue,moreover,to further clarify the clinical significance in diagnosis,treatment and curative effect evaluation for the breast cancer patients.Methods:145 cases of breast pathological archives,including 40 cases of fibroadenoma,90 cases of breast cancer and 15 cases of normal breast tissues(from the operation margin of more than 2cm)were collected in the Second Affiliated Hospital of Da Lian Medical University,which are all surgically removed from July to December 2015.Immunohistochemical streptavidin peroxidase method(S-P method)was used to detect the expression of UBE2 C and P53 in three different breast tissues,and to analyze the correlation between them and their relationship with clinicopathological characteristics in breast cancer.Results:(1)The expression of UBE2 C and p53 in 15 cases of adjacent normal tissues and 40 cases of breast fibroadenoma tissues were completely negative;UBE2C in breast cancer tissues was significantly overexpressed and its positive expression rate was 46.7%(42/90).The positive expression rate of p53 in breast cancer tissues was 26.7%(24/90).(2)The expression of UBE2 C in breast cancer tissues was related with lymph node metastasis(x~2 =8.601,P = 0.005),vascular or neural invasion(X~2=11.041,P = 0.001)and clinical stage((X~2=20.100,P = 0.000),but no significant relation was foud with the patient’s age,menopause,tumor size,degree of malignancy grade and the expression status of ER,PR,HER2(P > 0.05).The expression of p53 in breast cancer tissues was related with degree of malignancy grade(x~2 = 9.376,P = 0.009),ER(x~2 =41.644,P = 0.000),PR(x~2 =41.644,P = 0.000),HER2 expression status(x~2 =6.545 P = 0.016),but are not related with the patient’s age,whether menopause,tumor size,lymph node metastasis,clinical stage,vascular or nerve invasion(P > 0.05).(3).In 24 breast cancer patients with p53 positive expression,ube2 c positi ve expression rate was 75%(18 / 24)and in 66 breast cancer patients with p53 negative expression,ube2 c expression rate was 36.36%(36.);ube2c and p53 in the general breast cancer population has a certain not strong correlation(X~2=10.556,P = 0.002,r = 0.342).In the four kinds of of breast cancer with differe nt molecular classification,the positive expression rate of ube2 c are25.0%(Lumi nal A)、26.67%(Luminal B)、59.09%(HER2over-expression)and 88.89%(TN BC);the positive expression rate of ube2 c are 10.0%(Luminal A)、11.1%(L uminal B)、31.82%(HER2 over-expression)和 66.67%(TNBC).The expression of ube2 c and p53 both have significant statistical difference(UBE2C: X~2 =22.851,p = 0.000;P53: X~2 =20.019,p=0.000)in four kinds of of breast cancer with different molecular classification.The positive expression rate of ube2 c and p53 b oth were highest in TNBC,respectively is 80.5%(16 / 18)and 66.67%(12 / 18),significantly higher than that in other three types.UBE2 C and P53 were posi tively correlated in TNBC,with significant difference(X~2=9.990,P=0.006,r=0.527).Conclusions:(1)The positive expression of UBE2 C and P53 in breast cancer tissues were both obviously higher than that in the adjacent tissues and breast fibroadenoma tissues,which are mostly negative.(2)A certain correlation between between UBE2 C and P53 was found in the population of breast cancer,and they are positively correllated in the TNBC,and the statistical significance are marked.(3)Joint detection of UBE2 C and P53 expression is more helpful to the diagnosis,malignant degree evaluation,guiding treatment and curative effect prediction of breast cancer,etc.
