| Background and Objective: In 2010,there were 102900 new cases of colorectal cancer and 39670 cases of colorectal cancer in the United States,while 51370 patients died of colorectal cancer in the same year.Our country has large population base,the incidence was exactly far more than the United States.the morbidity and mortality showed an upward trend in recent years,with a high incidence in the elderly people.The pathogenesis is complex and may be related to many factors.without any symptoms in the early stage,it can not easily be found,In the late stage,the around invasion and distant metastasis of cancer can early occur.After radical resection,the prognosis is favorable,but there is no accurate indicator to monitor the prognosis of patients with colorectal cancer.In recent years,there are more and more researches respecting MSI(musashi)family.MSI family as RNA binding protein which is highly conserved in evolution.,with diversity of family expression and function,among which,Musashi2 protein plays an important role in cell asymmetric division,cell self-replication,aspect of self-regeneration capacity of cell.Meanwhile,by regulating translation process after gene transcription,MSI2 proteins maintain the degree of stem cell differentiation and self-renewal capacity,by regulating the Notch pathway,Wnt / β-catenin signal pathway embryonic directly or indirectly have some impacts on biological behavior such as stem cells,neural stem cells,hematopoietic stem cells and progenitor cells,germ-derived cells etc.In recent years,a growing number of experiments confirmed that MSI2 is also involved in the development process of gastric cancer,liver cancer,breast cancer,pancreatic cancer,nervous system cancer,leukemia,colorectal cancer and other cancers,and is closely linked with the Invasive and prognosis of cancer.By immunohistochemistry(SP),the study detect the expression of MSI2 which in colorectal cancer specimens and corresponding cancer tissues,and further analyze the relationship between MSI2 and clinical pathology features of colorectal cancer,to explore the relationship between MSI2 and the occurrence and development of colorectal cancerMethods: During March 2014----October 2015,there are 56 cases that are radical surgery after resection of colorectal cancer specimens and paired cancer tissues specimens from Liaoning Provincial People’s Hospital General Surgery.The cases use immunohistochemical SP method to detect the expression of MSI2 in specimens of colorectal cancer tissues and adjacent non-cancerous colorectal tissues,and analyze the relationship of clinical pathology features among gender,age,tumor location,lymph node metastasis,nerve and vascular invasion or not,Invasion degree,the level of preoperative serum CEA,the size of the tumor,the Differentiation of the tumor,TNM stage.Result: Forty-three cases appear high MSI2 expression with the rate of 85.7%(48/56)in colorectal cancer specimens,and 12 cases appear high MSI2 expression with the rate of 14.3%(12/56)in the adjacent non-cancerous colorectal tissues,using paired-samples T test,we find that there are obvious statistical difference between them(t=3.572,P=0.002).Using chi-square test,we proved the expression of MSI2 is irrelevant to patients ’ gender,age,tumor location,lymph node metastasis,nerve and vascular invasion or not,invasion degree,TNM stage,but is related to the size of the tumor,the differentiation of the tumor,the level of preoperative serum CEA.Conclusion: MSI2 emerges higher expression in colorectal cancer than in the adjacent non-cancerous colorectal tissues,and related to the size of the tumor,the differentiation of the tumor,the level of preoperative serum CEA.It shows that MSI2 is probably related to the disease of colon adenocarcinoma and has certain significance in further study on the occurrence and development of colorectal cancer,and may be new Aided Diagnosis of colorectal cancer and Corresponding therapeutic target. |
