OBJECT:Over the past 30 years, the incidence and mortality of gastric cancer rising steadily in China, gastric cancer is still in the anterior position of malignant tumors in the world. Therefore, early diagnosis and identification of gastric cancer is particularly important. In recent years, with the increased proportion of early gastric cancer, the 5 years survival rate of gastric cancer has been increasing.Traditional diagnostic methods of gastric cancer include barium meal, gastroscopy and biopsy. However, there are still some cases of gastric cancer diagnosised with poor prognosis, which is not only related to the discovery period, but also closely related to the molecular pathological types. Therefore,it is particularly necessary to find an early detection indicator or prognostic marker for gastric cancer. In this study, the expression of MSI2 gene and Ki67 in gastric cancer tissues and its relationship with clinical cases and prognosis of patients with gastric cancer were studied. We are committed to explore a prognostic predictor of gastric cancer and find a potential molecular target in gastric cancer.MATERIALS AND METHODS:This topic mainly includes two parts, which inlude clinical specimens and case data collection, gene expression detection in specimens and clinical follow-up of patients after surgery.Clinical specimens and case data collection:100 clinical specimens were collected and clinical data were recorded. The MSI2 gene expression in specimens was detected:Immunohistochemistry and qPCR were used to detect MSI2 gene expression in the collected specimens. The correlation between MSI2 gene expression and clinical data was analyzed.Postoperative follow-up:The survival curve was drawn according to the survival data, and the correlation between gene expression and prognosis was analyzed.RESULT:1.MSI2 gene is positively correlated with the pathological grade and differentiation of gastric cancer.2.The expression of MSI2 gene was positively correlated with that of Ki67 gene. The prognosis of patients with high MSI2 gene expression was worse than that of patients with low MSI2 expression. The difference was statistically significant.CONCLUSION:The increased expression of MSI2 gene in gastric cancer suggests that the prognosis of patients with gastric cancer is poor. The expression of MSI2 gene can be used as a potential molecular marker for the prognosis of patients with gastric cancer. It is expected to become a potential molecular target for the treatment of gastric cancer.
