Curcumin is the rhizome extracts of turmeric plant,which is a kind of natural linear diaryl heptane ketone compounds,as well as a kind of slightly acidic phenolics.Curcumin has various functions, such as anti-inflammation and anti-infection, adjust the dyslipidemia, regulating immune function, antioxidant, scavenging free radicals, and so on.With the deepening of the research on curcumin, finding that it may has great application value in the nervous system diseases, such as alzheimer's disease,Parkinson's disease, nerve pain and nervous system tumors, etc. However there are not much researches about curcumin in the mechanism of Parkinson's disease, especially for the mechanism of autophagy. The experiment was established by treating PC12 cells with MPP+ the classic neurotoxic drugs.This research curcumin could attenuate the MPP+-induced injury of PC12 through autophagy pathway.Through testing ROS level and the mitochondrial transmembrane potential, inquiry of curcumin on the regulation of autophagy and the protective effect of PC12 cells.Objective Through comparing injury group and curcumin group of cell survival rate, ROS production, mitochondrial membrane potential, and the expression level of LC3 II / I,p62. To investigate the protective effects of curcumin on the MPP+-induced injury of PC12 cells and the related mechanisms autophagy pathway.Methods The cell model of Parkinson's disease was established by treating PC12 cells with MPP+ for 24 h. The cell viability was determined by CCK-8 assay.The cellular ROS level and the mitochondrial transmembrane potential were measured by flowcytometry.The expression of LC3?/??P62 in PC12 cells were detected by Western blot.Results The MPP+ treatment reduced the viability of PC12 cells,which could be reversed by curcumin.And curcumin could inhibit the production of ROS and the disruption of mitochondrial transmembrane potential induced by MPP+.Moreover curcumin could reduce the expression of LC3?/??P62 in PC12 cells treated MPP+.Conclusion Curcumin could attenuate the MPP+-induced injury of PC12 through autophagy pathway.
