| Purpose: Replication in chronic heart failure rats model,through rat hemodynamic indexes and serum inflammatory cytokines tumor necrosis factor alpha(TNF-α)and interleukin 6(IL-6)content changes and myocardial pathological changes clearly Qiangxin Huoli Decoction on rats with chronic heart failure protection and on inflammatory response intervention effect,and further reveal the mechanism of action.Method:(1)The model of chronic heart failure in rats induced by intraperitoneal injection of adriamycin,according to body weight were randomly divided into blank control group,model group,reference group,Chinese medicine high,medium and low dose group.Carotid artery intubation and hemodynamic monitoring indicators;(2)Enzyme-linked immunosorbent adsorption method(ELISA)determination of the content of TNF-a and IL-6 in serum of rats and to observe the change of inflammatory reaction;(3)By pathological detection means of HE staining and Masson staining observation of heart failure rats myocardial state,determine the degree of myocardial injury and the effect of drug intervention.Result:(1)The experimental period,except for the blank control group,the other rats weight are different degrees of decline,and compared with model group,the hemodynamic index in each treatment group improved significantly.Among them,the effect of the high dose Chinese traditional medicine group significantly,and attached to the group had no significant difference;(2)By measuring the content of TNF-α and IL-6 in the serum of rats,we found that the high dose group of the Qiangxin Huoli Decoction can significantly reduce the level of the two factors,and the difference of the model group was significant(p<0.05);(3)Under the microscope observation of myocardial HE staining and Masson staining,the model group and the blank group according to the difference of apparent,the high dose group of the Qiangxin Huoli Decoction in improving the myocardial,inhibit myocardial fibrosis and myocyte edema effect is obvious.Conclusion: The vitality of the Qiangxin Huoli Decoction can by reducing the content of inflammatory cytokines TNF-α and IL-6 in the course of the development of the chronic heart failure to suppress the development trend of the inflammatory response and through the heart is strong,improve heart function,adjust the hemodynamic parameters,prevent myocardial hypertrophy,fibrosis and inhibit ventricular remodeling play preventive and therapeutic effect on chronic heart failure. |
