| Objective: Based on network pharmacology,the effective active ingredients of Qiangxin Decoction and its potential target and mechanism of action in the treatment of chronic heart failure were studied.Methods: The effective active ingredients and prediction targets of Qiangxin Decoction were mined by using TCMSP,and the information was standardized by Uni Prot database.The disease prediction target of "Chronic Heart Failure" was found by using five databases,including Gene Cards database,OMIM database,Pharmgkb database,TTD database,and Grug Bank database.After removing the duplicates,the disease prediction target was intersected with the drug prediction target,and the Wayne map was drawn.Then import the effective ingredients and intersection target data into the bioinformatics analysis software(Cytascape 3.8.2)to draw the "component-target-disease" regulatory network,and screen out the core components according to the degree.The interaction data of intersection target protein was obtained through STRING database,and the core target was obtained.Use R language to enrich and analyze intersection targets.Finally,the core components and core targets are verified by molecular docking.Results:(1)In this study,a total of 280 effective active ingredients of Qiangxin Decoction were screened,and 516 drug prediction targets were selected,among which the core components were quercetin and kaempferol.(2)Based on five major data,including Gene Cards database,OMIM database,Pharm GKB database,TTD database and Grug Bank database,a total of 2105 disease prediction targets of chronic heart failure were obtained,and 87 common targets of Qiangxin Decoction and chronic heart failure were obtained.(3)Based on STRING database to obtain the interaction data of intersection target proteins,the protein interaction network is drawn in the bioinformatics analysis software(Cytascape 3.8.2).Using the three topological analysis methods of Degree,Affinity and Betweenness in Cyto Hubba,it is found that proteins such as IL6,CASP3,VEGFA,EGFR,PPARG are the core target proteins of Qiangxin Decoction in the treatment of chronic heart failure.(3)There are 1692 GO function enrichment items and136 KEGG pathway enrichment items related to chronic heart failure,mainly involving biological processes such as wound healing,oxidative stress reaction,and signaling pathways such as lipid and atherosclerosis.(4)Through molecular docking of the core component and core target of Qiangxin Decoction,it was found that the binding energy between most receptors and ligands was less than or equal to-5 kcal/mol,with good binding activity and conformation.Conclusion(s): Qiangxin Decoction may act on core targets such as IL6 and CASP3 through core components such as quercetin and kaempferol,and participate in biological processes and signal pathways such as wound healing,oxidative stress,lipid and atherosclerosis,fluid shear stress,atherosclerosis,cell apoptosis,etc.to play a role in treating chronic heart failure.The biological process and related pathways of Qiangxin Tang in treating chronic heart failure are revealed to be very extensive,intervening in a large number of important pathological and physiological processes of chronic heart failure,providing objective basis for the multi pathway,multi target,and multi pathway pharmacological mechanism of Qiangxin Tang. |
