Objective: Discussion about the YouGui Capsule’s effect on Kidney-yang deficiency type of elderly infertility patients in the quality of oocytes, and intervention and regulation on JAK2,Cyclin D1 and SOD’s levels which are the critical and relative factors in JAK2/STAT3 pathway.To study the mechanism of elderly infertility women in JAK2/STAT3 pathway level and the effects and principles of traditional Chinese medicine of tonifying the Kidney in the process of treatment.Methods: In clinical research, 64 Kidney-yang deficiency type of elderly women undergoing IVF-ET were randomized into two groups, which were the YGC group, YGC combined with western medicine(32cases), and the placebo group, placebo combined with western medicine(32cases). In addition, 32 elderly women undergoing IVF-ET because of male factors were randomized into the normal control group, placebo combined with western medicine(32cases). The syndrome scores of Kidney-yang deficiency between the fist two groups were evaluated, and the days and amount of Gn, serum level of E2,P,LH on HCG injection day, the number of oocytes, high quality oocyte rate,fertilization rate, the number of embryo, clinical pregnancy rate and the level of SOD, JAK2 and Cyclin D1 which are the relative and critical factors in JAK2/STAT3 pathway of all groups were monitored.Results: 1. Symptoms of Kidney-yang deficiency in YGC group after treatment was significantly improved(P<0.01), while the placebo group’s was little changed(P>0.05). The scores on HCG injection day between the two groups and the difference of scores before and after treatment in the two groups were statistically significant(P<0.01). 2. The days and amount of Gn, level of E2 on HCG injection day, the number of oocytes, high quality oocyte rate, fertilization rate, the number of embryo and clinical pregnancy rate of three groups were statistically significant(P<0.05 or <0.01).Compared with placebo group, YGC and normal control group had fewer days and lower amount of Gn, higher level of E2, number of oocytes, high quality oocyte rate,fertilization rate and number of embryo(P<0.05 or <0.017 or <0.01),while there was no big difference between YGC group and normal control group(P>0.05 or > 0.017). Clinical pregnancy rate in YGC group and normal control group was higher than that in placebo group(P<0.025).The level of P and LH made no significant distinction(P>0.05). 3. By analyzing the level of JAK2, Cyclin D1 and SOD which are the critical and relative factors in JAK2/STAT3 pathway of all groups, we found that it was statistically significant(P<0.05).The content of SOD, JAK2 and Cyclin D1 in YGC and normal control group was higher than that in placebo group, the difference was statistically significant(P<0.05 or <0.01),while there was no big distinction between YGC group and normal control group(P>0.05).Conclusion: 1. YouGui Capsule could obviously improve symptoms of Kidney-yang deficiency, the number of oocytes, high quality oocyte rate, fertilization rate, the number of embryo and clinical pregnancy rate. 2. You Gui Capsule may improve the outcomes of Kidney-yang deficiency type of elderly infertility female by increasing and regulating the expression of SOD,JAK2 and Cyclin D1 which are affiliated and relative to JAK2/STAT3 pathway. 3. Based on JAK2/STAT3 pathway, expressing the mechanism of “promote reproduction by tonifying kidney” and providing a more objective basis for the theory of Kidney Controlling Reproductivity in TCM in the view of genomic-pathway-proteomics.
