| Objective:To investigate the effects of six Dihuang particles on quality of oocyte in patients with kidney yin deficiency type of infertility and mechanism of FADD and Caspase-3 in Fas signal transduction pathway to improve the symptom of Kidney yin deficiency syndrome and restrain the apoptosis of ovarial granulosa cells. Studying of six Dihuang particles on the levels of possible mechanism may be in the Fas signal transduction pathway level and Key protein FADDMethods:a total of 78 infertility patients with deficiency of kidney in vitro fertilization and embryo transplantation in the treatment of patients, Randomly divided into six Dihuang particles group and placebo group(treatment the treatment group take six Dihuang particles combined with routine IVF treatment in the follicular phase, the placebo group and normal control group received a placebo and follicular phase combined with routine IVF in the treatment of kidney deficiency, kidney yin deficiency syndrome integral change of infertility patients; Two groups were treated with three menstrual cycle. Groups were compared in patients with kidney yin deficiency syndrome integral change, hormone dosage and duration of medication, injection of HCG on serum E2, LH, P levels, the number of oocytes retrieved, high-quality oocytes rate, fertilization rate, good quality embryo rate. each group were collected on the day of oocyte retrieval in mature human granulosa cell, Determination of FADD, Caspase-3 protein by Western blot method.Results: 1.Before and after treatment in treatment group compared to decyl kidney yin deficiency syndrome integral, the difference was statistically significant(P<0.05); before and after placebo treatment of kidney yin deficiency syndrome integral compared, no significant difference(P>0.05); patients in the treatment group of oocytes and fertilization rate high quality egg rate, good quality embryo rate, pregnancy rate, biochemical and clinical pregnancy rate higher than the placebo group, the difference was significant(P<0.05); 2 It was found that the treatment group expression of FADD, caspase-3 were lower than those in the placebo group, the difference was significant(P<0.05).Conclusion: 1.Six Ingredient Rehmannia particles can obviously improve patients with kidney yin deficiency type of infertility due to kidney deficiency state, improve the quality of egg rate, good quality embryo rate, improve the clinical pregnancy rate. 2.Six Ingredient Rehmannia particles may be through Fas signal transduction pathway, factor FADD expression down regulated, thereby restraining granulosa cell apoptosis, so as to improve the kidney deficiency patients with infertility IVF outcome; 3.Based on reducing the FADD on Fas signal transduction pathway to interpret "mechanism of nourishing kidney promote reproduction", provide a more objective basis for "the kidney controlling reproduction" theory. |
