| Objectives Retrospective analysis of serum lipid levels from health check-up population for the improvement of Cholesterol Management in Tangshan City. The investigation on the availability of circulating mi R-28-5p as a tool for HDL-C stratification management in healthy subjects.Methods 1 The data of 25, 197 subjects, including 11114 males and 14083 females, were collected from 2009 to 2014 in Health Check-up department in Tangshan Gongren Hospital after filterd the abnormal data based on liver injury, abnormal blood glucose,kidney injury and disease history. Lipid profile were analyzed with R 3.0.2. 2 We enrolled174 healthy subjects with normal TC, LDL-C and TG levels. The plasma was isolated by two-step centrifugation method and total RNA was purified by Ambion PRISM mi RNA isolation kit. The mi RNAs, mi R-28-5p and mi R-191-5p, in plasma were quantified using Taq MAN stem-loop mi RNA Real-time quantitative PCR. The expression level of mi R-28-5p was normalized to control reference, mi R-191-5p, and 2-?Ct was reported as the relative expression level. The data was analyzed by Graph PAD Prism 5.0.Results 1 Levels of TC, TG, LDL-C and HDL-C in healthy subjects were 4.85(4.28,5.29), 1.18(0.86,1.68), 2.73(2.21,2.79) and 1.18(1.00,1.40) respectively. Each lipid index has significant difference in gender. TC levels in male were higher then female in30-39 and 40-49 group; TG, LDL-C levels in male were higher than female in 20-29, 30-39, and 40-49 group; HDL-C levels in male were lower than female in each group based on age. The median level of TC had a increasing trendence with age from 4.02 mmol/L up to 5.06 mmol/L in male and from 4.40 mmol/L up to 5.57 mmol/L in female. The median level of TG in male increased from 1.16 mmol/L up to 1.45 mmol/L of 40-49 years and then decreased to 1.20 mmol/L, while its levels in female increased from 0.89 mmol/L up to 1.47 mmol/L. The median level of LDL-C had a increasing trendence from 2.57 mmol/L up to 3.03 mmol/L in male and from 2.28 mmol/L up to 3.13 mmol/L in female. The median level of HDL-C increased from 1.06 mmol/L up to 1.11 mmol/L with age in male,but not in female. Total prevalence of dyslipidemia was 42.48% among this population.The prevalence of high TC, high TG, high LDL-C, low HDL-C were 8.87%, 11.97%,6.04% and 29.69% respectively. The prevalence of Low HDL-C dyslipidemia in male and female was 42.94% and 19.23%, which was higher than other abnormal indexes. The prevalence of High TG, high LDL-C and high TC was 16.77%, 8.00% and 6.98% in male,and 8.19%, 9.52% and 5.29% in female, respectively. 2 The relative expression level of circulating mi R-28-5p in male and female was 0.003386 2-?Ct and 0.004247 2-?Ct,respectively. There was significant difference between male and female(W=4263, P=0.02794).Conclusions 1 The population with Low HDL-C dyslipidemia is major objective for Cholesterol management and CHD preventation in Tangshan City. 2 Circulating mi R-28-5p is not suitable as a indicator for the stratification of HDL-C level. |
