?Objective?To investigate PTTG1 promoter Cp G island methylation rate in tumor tissues and adjacent tissues of the cancer in the patients with sporadic colorectal carcinoma and further research the relationship between the pathological and clinical.?Methods?Bisulfite sequencing polymerase chain reaction(BSP) was detected in sequencing method of 40 patients with colorectal carcinoma tissues and adjacent tissues PTTG1 gene promoter region Cp G island(-1~-400 bp bp) within the scope of the bases of methylation rate; Western Blot(WB) and real-time fluorescent quantitative polymerase chain reaction(q RT- PCR) technology PTTG1 protein and m RNA expression was detected in case; And further analysis of the relationship between expression and pathological features and clinical.?Results?1. The result shows that PTTG1 gene promoter Cp G island(-1~-400bp) has a total of 32 methylation site. Carcinoma tissue of PTTG1 promoter Cp G island methylation rate is 19.05 ± 4.45.For the tissue adjacent to carcinoma groups methylation rate is 68.07 ± 3.71.Both comparison difference has statistics meaning(P < 0.05). Colorectal cancer tissue PTTG1 gene promoter region of the Cp G island methylation rate is lower than the tissue adjacent to carcinoma, and the methylation rate of cancerous tissues and is associated with the tumor staging,and the depth of serosal invasion. But wasn't correlated with patients' age?gender, the size of tumor and the degree of lymph node metastasis; 2. The expression of PTTG1 protein in colorectal carcinoma tissues is 2.79±0.23.For the tissue adjacent to carcinoma groups is 1.00±0.17.The difference was statistically significant( P < 0.01). Colorectal carcinoma tissue PTTG1 protein showed high expression to the tissue adjacent to carcinoma. The expression of PTTG1 gene in colorectal cancer tissue rate is associated with the depth of serosal invasion and the tumor staging. But wasn't correlated with patients' age?gender, the size of tumor and the degree of lymph node metastasis; 3. q RT-PCR results showed that the PTTG1 m RNA level of colorectal carcinoma tissues was significantly higher than that of cancer adjacent tissues(P=0.028).?Conclusions?1. PTTG1 promoter region within the scope of-1~-400 bp cancerous tissue Cp G island methylation rate is lower than the tissue adjacent to carcinoma; 2. The expression of PTTG1 in tumor tissues is higher than adjacent carcinoma tissues, The higher expression is associated with the depth of serosal invasion. and the tumor staging. May become the etiology in patients with colorectal cancer screening, prognosis judgement, provides the basis.
