Objective: To detect the expression of RB and mutant type p53 in breast cancer tissues, analyze the relationship between the expression of RB and mutant type p53 with the clinicopathological features. Then clarify whether expression of RB and mutant type p53 genes could be prognostic factors in breast cancer patients.Methods: 140 cases of breast carcinoma specimens which were resected by surgical operation in Wuhan Union Hospital from January 2001 to August 2004 were collected. All the patients received proper treatment. The expression of RB and mutant type p53 were detected by tissue microarray technology and immunohistochemical staining method. SPSS 22.0 was used to analyze the relationship among the expression of RB and mutant type p53 with age, location of tumor, phase of tumor, histophathologic grade, status of lymph nodes metastasis, etc. Then assessed the prognosis value of the expression of RB and mutant type p53 in breast cancer patients.Results: Lower RB expression was related to status of lymph nodes metastasis, expression of ER, expression of Her-2 and subtypes of breast cancer(P<0.05). Higher mutant type p53 expression was related to expression of ER and subtypes of breast cancer(P<0.05). The expression of RB in TNBC was lower than others(P=0.004).The expression of mutant type p53 in TNBC was higher than others(P=0.003).There was more cases in which combined with low RB and high mutant type p53 expression in TNBC than others(P=0.001). Patients in whom RB expression was low received shorter overall survival compared with the high ones. The combination of mutant type p53 and RB expression revealed that the cases with low RB and high mutant type p53 expression had significantly shorter overall survival, but this combination did not show an independent prognostic factor for survival of breast cancer by univariate analysis.Conclusions: The combination of low RB expression and high mutant type p53 expression revealed more cases in TNBC compared with other groups. And got shorter overall survival than others. Though this group is not an independent prognostic factor for survival of breast carcinoma, but it could be regarded as a prognostic indicator in breast cancer, and provide theoretical basis for the gene therapy of breast cancer.
