Sperm is the key productive cell of mammals. Plenty of evidence suggests that sperm spercific and pH dependent CatSper and KSper are two main ion channels in sperm, which is also the key factor decides the fertilization ability of sperm. Although in recent years with the classic patchclamp technique important progress has been made in studies of sperm ion channels, sperm characteristics of small volume limited the patchclamp technique to be widely used in the study of sperm.It is a key way to study cell's response to environmental stimuli and screen cellular functions' s regulationg candidate in the simulation environment by bulding a cell electrophysiological model with compose and character known channels in cells. Simulation software CeLwhich is developed by our lab is applied in this arcle combining with the existing CatSper and KSper electrophysiological data and anelectrophysiological model is built which can match the experiment data of pH dependent membrane potential very well. Applying this model, we found that the sealing resistance has significant effects on mice sperm membrane potential, which provides the basis to understand why different membrane potential is recorded from diferent sperm. The wild type and Slo3 knockout mice sperm alkality fluorescence experiment and simulation comparison proved Slo3 affected the CatSper channels by dreving force for the CatSper action rather than the chemical coupling between two channels or other interactions. In addition, with the application of the model we can simulate ion channel inhibitors effects on sperm intracellular calcium concentration, and concluded that the block ratio of CatSper and KSper by inhibitors.This paper provides an effective altemative method for the study of sperm function regulating mechanism, which is particularly suitable for evaluation of channel inhibitors or activators influence on sperm functions and has a good application potential for the research of sperm function regulating mechanism and the development of male contraceptives.
