Objective: To study the relationship between the isolated systolic hypertension(ISH) and the gene polymorphisms of homocysteine metabolism-related enzymes which incloud MTHFR C677 T, MTRR A66 G, MS A2756 G.Methods: 220 patientswith isolated systolic hypertension from tianjin chest hospital outpatient clinic were collected as the experimental group and 130 patients without high blood pressure were collected as the control group. The levels of homocysteine,total cholesterol(TC), triglycerides(TG), low density lipoprotein(LDL), c-reactive protein levels(hs-CPR) and uric acid levels in their blood were detected and the gene polymorphisms of MTHFR C677 T, MTRR A66 G, MS A2756 G were tested by using the methods of molecular biology. Finally,SPSS18 statistical software were used to analyze the relationship between isolated systolic hypertension and homocysteine metabolism enzyme gene polymorphism.Results:1.The comparation of group general information between ISH group with control group general information, The gender, age, smoking ratio of the two groups had no obvious difference, but the morbidity of diabetes mellitus in ISH group were higher than the control group(P < 0.05),and so as the homocysteine levels and c-reactive protein levels(P < 0.05).2.The Logistic regression analysis were applied by regard ISH as dependent variables.than it is concluded that homocysteine levels, age, uric acid levels were the independent risk factors of the disease(P < 0.05), while gender, smoking status, level of total cholesterol, triglycerides, low density lipoprotein levels, hypersensitive c-reactive protein were not.(P > 0.05).3. The Hcy levels of male were higher than women, and it had statistical significance(P < 0.01).No matter in in ISH group or in the control group.and In men and women in the crowd, Hcy levels in ISH group were more than control group in both men group and women group( P < 0.01).4. The study found there was no linear relationship between Hcy levels and age.5. ISH group and control group in MTHFR gene C677 T, MTR A66 G, MS gene A2756 G various genotype frequency distribution were all accord with Hardy- Weinberg equilibrium test(P > 0.05), which showed that the ISH group and the control group belong to genetic balance, and could be reached as the group representative. 6. MTHFR gene C677 T, The C/T, T/T genotype frequency in ISH group were greater than the control group(P < 0.05), and the T allele frequency in ISH group were also higher than the control group(P < 0.05).In gene MTRR A66 G, The frequency of ISH group A/G, G/G and G allele is lower than the control group, but it may be because of the small sample sizes, no significant statistical difference significance were found, In this study,we could not find obvious difference between the two groups in the MS gene polymorphism.7.Between different genotypes in MTHFR C677 T, MTRR A66 G, MSA2756 G.homocysteine levels, low density lipoprotein, cholesterol levels, hypersensitive c-reactive protein level, uric acid level, age, were observed no significant difference(P > 0.05).Conclusions:1. The homocysteine levels, hypersensitive c-reactive protein levels and the prevalence of diabetes in ISH group were higher than the control group, they may be the certain promoting role in the pathogenesis of ISH.2. Homocysteine levels, age, uric acid levels were the independent risk factors of ISH, the increasing of homocysteine levels, age, and blood uric acid levels may increased the risk of ISH.3. The homocysteine levels of male crowd were higher than that of women, but it had nothing to do with whether suffering from hypertension..4. Age had be found no obvious relation with homocysteine levels. 5. The mutation of C677 T in MTHFR gene may be associated with the incidence of ISH.while the MTRR A66 G gene and the MS A2756 G gene polymorphism may be not the correlative genes of ISH.
