Prescription of Jujube benevolence nerves (PJBN) is often used in the treatment of insomnia and other chronic diseases, to zizyphus jujube kernel as monarch drug, and the compatibility of fructus schisandrae vinegar and Salvia. This topic based on the theory of compatibility of traditional Chinese medicine, selection of Spinosin and ferulic acid as the indicators, using high performance liquid chromatography, to detect tissue distribution of Spinosin and ferulic acid group, Jujube kernel group, Jujube kernel-Fructus schisandrae group, Jujube kernel-Salvia miltiorrhiza group and PJBN group in normal rats. The results were as follows:1. Method of spinosin and ferulic acid in the optimization of HPLC for the determination of drug in zizyphus jujube kernel. Results show that the method of strong specificity, good linear relationship. In order to provide an experimental basis for further study of the pharmacokinetics of the drug.2. Set up a HPLC analysis method which used to analysis spinosin and ferulic acid in rat tissue samples. The results showed that, spinosin and ferulic acid in the detection range has a good linear relationship, endogenous substances in the tissues and measured components did not interfere with. High, medium and low concentrations of the spinosin and ferulic acid tissue sample recovery rate was above 90%, precision, stability and other indicators of the RSD values were less than 10%, in line with the requirements of biological sample analysis.3. Study on the distribution of PJBN extracted liquid in rat tissues. Results showed that, Spinosin and ferulic acid can penetrate the blood-brain barrier into the cerebrospinal fluid play a pharmacodynamics, in the brain tissue of the earliest detectable spinosin is 0.25h, ferulic acid is 1h; A higher concentration in the heart, liver and kidney tissue. According to reports, this may be related to the theory of traditional Chinese medicine Suanzaoren meridian has certain relations theory. It may enhance its sedative effect on the central nervous system.4. Compatibility on zaorenanshen side effects of tissue distribution. Results showed that, compared with Jujube kernel group, Jujube kernel-Salvia miltiorrhiza group Spinosin and ferulic acid in the peak concentration increased, that explains some chemical components in Salvia miltiorrhiza promotes spinosin and ferulic acid in vivo absorption, distribution; Jujube kernel-Fructus schisandrae group Spinosin in the peak concentration decreased, that explains some chemical components in Fructus schisandrae reduce spinosin in vivo distribution; PJBN group spinosin and ferulic acid in the brain distribution significantly increased, indicating that the prescription can improve the effective concentration in rat tissues. Therefore, we speculate that spinosin and ferulic acid may be effective sedative and hypnotic effects of Jujube kernel based central pharmacodynamic material. The experimental results further improve the PJBN in vivo pharmacokinetic processes, and it provided an effective reference for the study of the mechanism of compatibility and the material basis of drug action.
