Study On The Chemical Basis And Pharmacokinetics Of Shuang HuangLian

Shuang Huang Lian (SHL) is a classical Chinese traditional medicine compound for cold-pungent diaphoresis, heat-clearing and detoxification. It is simple to compose the prescription, quite effective, and widely used in clinical practice. In this paper, three different formulations of SHL preparation product are chosen for the study on the pharmacokinetics, aiming to provide a scientific basis for the establishment of the quality control system and the clinical dosing regimen.This study is based on the overall view of Chinese medicine. A method that combines finger printing and multiple components pharmacokinetic is tried to study the pharmacokinetics characteristics of SHL. The finger printing is used to explore the material basis of SHL, and to compare the chemical component differences of the three different formulations of SHL preparation:the oral liquid, the capsule, and the powder injection agent. And then the pharmacokinetics of three different formulations of SHL are studied and compared. The differences and correlation of multiple components physiological disposition are analyzed. The labeled components that may character the physiological disposition of SHL are identified. All above work are so as to provide a theoretical basis for the establishment of quality control system for the preparation of SHL.The contents in this paper include the following two parts:1. The study on the chemical components of the three formulations of SHL preparation product. In this study, the HPLC is adopted to analysis the fingerprint chromatogram of chemical components of the three different formulations:SHL capsules, SHL oral liquid and SHL powder injection agent, to specify their common peaks, and to analyze the total peak area of the PC A.The HPLC-Q-TOF-MS is used to confirm CA, PHIL, BCL, BCN, WGN, and the HPLC is used for the contents determination of them. And the contents of these compounds in the three different formulations are compared. The results show that the similarities of the chemical components of the three formulations are very high, but they have a major difference in content of their main chemical components.2. The study on pharmacokinetics of the three different formulations of SHL.In this study, the pharmacokinetic activities of BCL and BCN in the SHL soft capsule, oral liquid and powder injection agent under different dosages are compared. The differences of the vitro metabolism process and the interactions between the components are analyzed. The results show that, BCL has different pharmacokinetic characteristic in different administration pathway. The similarities of pharmacokinetic characteristics of different formulations in the same administration pathway are high. These provide a scientific basis for the clinical administration regimen of SHL.
