| Objective:To investigate the expression of ORMDL3(orosomucoid 1-like 3)single nucleotide polymorphisms(SNPs)in the pathogenesis of asthma in children with mycoplasma pneumonia(MP).Methods:A total of 200 patients with MP infection were enrolled in Shandong Provincial Affiliated Provincial Hospital from January 2015 to December 2015.The follow-up observation was 1 year,Repeated or persistent coughing and/or wheezing were diagnsed asthma and named MP-asthma group no repeated or persistent cough and/or wheezing were named MP-non-asthma group.Collect general information about selected children,clinical manifestations,personal and family history of allergies,Chest X-ray and lung function,MP-IgM,MP-RNA and so on.Two SNPs loci rs4794820 and rs7216389 were selected from ORMDL3 gene.Genomic DNA of peripheral blood sera extraction was performed using a Tguide S32 Nucleic Acid Extractor.The above loci of each group were genotyped by FLUIDIGM system.SPSS 19.0 software was used for statistical analysis,and their distribution differences were compared in each group.Results:A total of 200 cases of MP infection were hospitalized,6 cases were lost,and 194 cases were included in the study.Among them,136 were male and 58 were female.The male was about 2.3 times of that of female.The minimum age was 3 years and the maximum age was 12 years.Among the 194 cases,the patients who developed asthma were MP-asthma group(63 cases),and no recurrent or persistent coughing and wheezing were MP-non-asthma group(131 cases).The proportion of asthma developed after MP infection was about 32.5%(34/194).2.There was no significant difference in sex and age distribution between MP-asthma group and MP-non-asthma group(P>0.05).3.ORGDL3 gene loci rs4794820 were AG,GG,AA.The frequency of homozygous GG genotype was highest in rs4794820 locus in MP-asthma group and MP-non asthma group(60.3%vs 42.7%,respectively),followed by heterozygous AG genotype(34.9%vs 38.2%,respectively).The lowest genotype was homozygous AA genotype(4.8%vs 19.1%,respectively).Compared with genotype and allele frequency of rs4794820 in MP-asthma group and MP-non asthma group were significantly different.The difference was statistically significant.(P<0.05).(respectively,x2=8.877,P=0.012;x2=9.786,P=0.002).The The frequency of GG homozygous genotype and G allele in MP-asthma group was significantly higher than that in MP-non-asthma group(60.3%和 42.7%,78.8%和 61.8%,respectively,P<0.05).The frequency of AA homozygous genotype was the lowest among the two groups,but was significantly higher in the MP-non-asthma group than in the MP-asthma group(19.1%和 4.8%,respectively,P<0.05).The difference was statistically significant.4.ORGDL3 gene loci rs7216389 were TT,TC,CC,The frequency of homozygous TT genotype was highest in rs7216389 locus in MP-asthma group and MP-non asthma group(65.1%vs45%,respectively),followed by heterozygous TC genotype(28.6%vs 38.9%,respectively).The lowest genotype was homozygous CC genotype(6.3%vs16.0%,respectively).Compared with genotype and allele frequency of rs7216389 in MP-asthma group and MP-non asthma group were significantly different.The difference was statistically significant.(P<0.05(respectively,x2=12.134,P=0.002;x2=8.837,P=0.003).The frequency of TT homozygous genotype and T allele in MP-asthma group was significantly higher than that in MP-non-asthma group(65.1%vs45.0%,79.4%vs64.5%,respectively,P<0.05).The frequency of CC homozygous genotype was the lowest among the two groups,but was significantly higher in the MP-non-asthma group than in the MP-asthma group(16.0%vs6.3%,respectively,P<0.05).The difference was statistically significant.5.ORGDL3 rs4794820 GG genotype and rs7216389 TT genotype have the risk of asthma,(respectively,OR=2.036,95%CI=1.104~3.754,χ2=5.258,P=0.022;OR=2.274,95%CI=1.221~4.236,x2=6.841,P=0.009,).The difference was statistically significant(p<0.05).6,The ratio of serum IgE raising in MP-asthma group was higher than that in MP-non-asthma group,There was significant difference(P<0.05).7.The rs4794820 and rs7216389 genotype AG/TC FEV1/FVC%and FEV1 in the MP--asthma group were lower than genotype GG+TT,and there was significant difference(p<0.05)。CONCLUSION:1.ORMDL3 is associated with asthma induced by MP infection in children,and increased risk of asthma in children with MP infection.2.The SNPs rs4794820 GG genotype and rs7216389 TT genotype were the risk gene for asthma associated with childhood MP infection,And s4794820 AA genotype and rs7216389 CC genotype may be the protective gene.3.Elevated serum IgE levels and decreased pulmonary function tests after MP infection can be a risk factor for MP infection-related asthma. |
