| Objective:In order to investigate the pathophysiological mechanism of Ras gene mediated gastric precancerous lesions(PLGC)in rats,and use of traditional Chinese medicine compound modified qifangweitong granules on PLGC model rats to intervene.Explore modified qifangweitong granules of PLGC rats the intervention effect of Ras gene and protein,and modified qifangweitong granules on the mechanism of action of PLGC,to provide the experimental basis for the exploration of traditional Chinese medicine modified qifangweitong granules in the treatment of PLGC,but also for its extension to provide a theoretical basis for clinical application.Methods:The 60 male rats were randomly divided into four groups,blank group,model group,Weifuchun group ang modified Qifangweitong granules group,each group only 15.In addition to the blank group were given tap-water,the rest three groups were given 180 mu g/ml in the concentration of N-methyl-N ’-nitro-N-liquid nitrosoguanidine(MNNG)free drink for three months,after the successful model of PLGC rats,blank group and model group rats were given saline by gavage every day once,and Weifuchun group and the modified Qifangweitong granules group daily were given Weifuchun aqueous solution and modifiedQifangweitong granules in aqueous solution by gavage once.Continuous lavage treatment groups of rats after 30 days,24 hours in a fast,under anesthesia,profile control in stomach tissue,under the light microscopy observation of rat gastric mucosal tissue pathological changes,and using modern molecular technology of real-time fluorescent quantitative polymerase chain reaction(RT-PCR),immunohistochemistry and Western Blot method(Western Blot)testing of each rat gastric mucosal tissue Ras gene and protein expression.Results:(1)Through the light microscope to observe the pathological changes of gastric mucosa in rats: compared with the blank group,model group,gastric mucosal inflammation in rats with severe infiltration,mucosal thinning,gland atrophy and irregular arrangement,visible intestinal metaplasia and atypical hyperplasia(p<0.01);Compared with the model group,Weifuchun group and modified Qifangweitong granules in rat gastric mucosa tissue precancerous lesions had significantly improved(p<0.01);Weifuchun group and modified Qifangweitong granules group compared the degree of gastric mucosal precancerous lesions of rats(p>0.05).(2)The expression of Ras gene in gastric mucosa of rats in each group was detected by real-time PCR technique.The results showed that the expression of Ras gene was significantly lower than that of model group(p<0.01)in the treatment group and the combination of the modified Qifangweitong granules groups.(3)The expression of Ras protein in gastric mucosa of rats in each group was detected by immunohistochemistry and Western Blot.The expression of Ras protein in the gastric mucosa of the rats in the treatment group and the rats in the modified Qifangweitong granules group was significantly lower than that in the model group(p<0.01).Conclusions: 1.The modified Qifangweitong granules can reduce the degree of infiltration of stomach of PLGC rat gastric mucosal inflammation cells,improve or even reverse the gland atrophy,intestinal metaplasia and atypical hyperplasia precancerous lesions.2.By modified Qifangweitong granules by Ras gene expression and protein expression of PLGC in gastric mucosa of rats,differentiation and proliferation of normal and orderly control cells,inhibit cancer cell proliferation,and induce cancer cell apoptosis and promote its development to the direction of apoptosis,which may be modified Qifangweitong granules block or reverse PLGC to one of the mechanisms of gastric cancer development.3.Modified qifangweitong granules through additional integrity on the one hand,improve the body’s immune system,thus enhancing the ability of gastric mucosal injury and repair,as well as defense;On the other hand by clearing away heat、toxic materia and dampness,inhibiting the growth of Helicobacter pylori infection,blocking the development of gastric precancerous lesions from the cause;in addition,the qi stagnation,phlegm dampness,blood stasis,qi stagnation,phlegm,clearing function of congestion as pathological products,improve gastric mucosal microcirculation,promote the absorption of inflammation.Enhanced mucosal injury repair and defense. |
